PENETAPAN KADAR METAMPIRON DAN FENILBUTAZON DALAM SEDIAAN KAPSUL SECARA SPEKTROFOTOMETRI
DERIVATIF DENGAN METODE ZERO CROSSING ABSTRAK
Campuran metampiron dan fenilbutazon merupakan salah satu jenis kombinasi dalam sediaan kapsul. Penetapan kadar kapsul metampiron dan fenilbutazon tidak terdapat dalam monografi, baik pada Farmakope Indonesia edisi V (2014) maupun United States Pharmacopeia (USP) edisi XXX (2007) sehingga diperlukan suatu metode analisis yang memenuhi uji validitas pada penetapan kadarnya. Tujuan penelitian ini adalah untuk menguji validasi metode spektrofotometri derivatif dalam menetapkan kadar campuran metampiron dan fenilbutazon dalam sediaan kapsul.
Metode penelitian yang dilakukan adalah pengambilan sampel secara purposif terhadap campuran metampiron dan fenilbutazon pada sampel sediaan kapsul X serta penetapan kadar secara spektrofotometri derivatif metode zero
crossing dalam pelarut Etanol-HCl 0,1N.
Hasil penelitian menunjukkan bahwa kadar metampiron pada sediaan kapsul X sebesar 100,57% dan kadar fenilbutazon sebesar 99,08%. Dari hasil analisis penetapan kadar sampel campuran metampiron dan fenilbutazon dalam sediaan kapsul menunjukkan baik metampiron maupun fenilbutazon memenuhi persyaratan kadar untuk sediaan kapsul metampiron tunggal menurut USP edisi XXX (2007) dan fenilbutazon memenuhi persyaratan kadar tunggal menurut Farmakope Indonesia edisi V (2014). Hasil uji validasi yang dilakukan terhadap sediaan kapsul X, diperolehan persen kembali dari metampiron adalah 99,673% dengan simpangan baku relatif (RSD) = 1,33% dan untuk fenilbutazon diperoleh persen perolehan kembali sebesar 99,59% dengan simpangan baku relatif (RSD) =1,23%.
Berdasarkan hasil penelitian yang dilakukan, dapat disimpulkan bahwa metode spektrofotometri derivatif dengan pelarut Etanol-HCl 0,1 N dapat diukur pada derivat kedua dengan panjang gelombang analisis 251,20 nm untuk metampiron dan 222,60 nm untuk fenilbutazon. Serta memenuhi persyaratan akurasi dan presisi, dan dapat digunakan untuk menetapkan kadar metampiron dan fenilbutazon pada sediaan kapsul.

DETERMINATION OF METHAMPYRONE AND PHENYLBUTAZONE IN CAPSULES SUPPLY BY DERIVATIVE SPECTROFOTOMETRY
WITH ZERO CROSSING METHOD ABSTRACT
The compound of methampyrone and phenylbutazone is one of combination in capsule supply. Determination of content of methampyrone and phenylbutazone in capsul did not found in monography, either in the fifth edition Indonesian Pharmacopeia (2014) or United States Pharmacopeia (USP) 30th edition (2007) that requires an analysis method that meets the test of validity in determining the content. The aim of this study was to test the validation of derivative spectrophotometry method in determination the content methampyrone and phenylbutazone in capsule.
The method of this research was done by purposive sampling to methampyrone and phenylbutazone mixture of the sample capsule X content using derivative spectrophotometry with zero crossing technique and determination in Ethanol-HCl 0.1 N.
The research results were obtained the methampyrone and phenylbutazone content of the sample capsule X 100.57% for methampyrone and phenylbutazone 99.08%. Based on the results of analysis determine the sample content of methampyrone and phenylbutazone compound in capsule supply methampyrone fulfilled the requirements in USP 30th edition (2007) and phenylbutazone fulfilled the requirement of capsule in the fifth edition Indonesian Pharmacopeia (2014). The results of validation test on the capsule X, the percent recovery for methampyrone is 99.673%, relative standard deviation (RSD) = 1.33% and for phenylbutazone, the percent recovery is 99.59% with relative standard deviation (RSD) = 1.23%.
Based on the results of research, that derivative spectrophotometry method with Ethanol-HCl 0.1 N and measured at the second derivate with wavelength of 251.20 nm for methampyrone and 222.60 nm for phenylbutazone. And fulfilled the requirements of accuracy and precision and can be used to determinate the content of methampyrone and phenylbutazone in capsule.
