Pembuatan Reagen Bradford
Commasive blue
sebanyak 0,01 gram dilarutkan kedalam 50 ml etanol 95% dan
ditambah dengan asam fosfor 85% sebanyak 100 ml. Larutan yang digunakan
diencerkan dengan menggunakan ddH
2O hingga 1000 ml. Larutan tersebut kemudian di
saring dan disimpan dalam botol gelap pada suhu rendah (Bollag & Stuart, 1991).

Pembuatan Kurva Standar Protein
Bovine Serum Albumin
(BSA) sebanyak 100 mg dilarutkan kedalam 100 ml ddH
2O
sehingga diperoleh larutan stok BSA 1 mg/ml. Selanjutnya dibuat seri pengenceran
larutan standar dengan konsentrasi 0 mg/ml; 0,1 mg/ml; 0,2 mg/ml; 0,3 mg/ml; 0,4
mg/ml; 0,5 mg/ml; 0,6 mg/ml; 0,7 mg/ml; 0,8 mg/ml; 0,9 mg/ml; dan 1 mg/ml. Dari
masing-masing konsentrasi diambil 0,1 ml larutan standar dan ditambahkan 5 ml larutan
Bradford. Kemudian didiamkan selama 30 menit dan diukur absorbansinya pada λ 595
