UJI AKTIVITAS ANTIBAKTERI DARI HASIL OPTIMASI
TRANSESTERIFIKASI MINYAK KELAPA SAWIT (Elaeis guineensis Jacq)
ABSTRAK
Minyak kelapa sawit (Elaeis guineensis Jacq.) merupakan salah satu komoditas pertanian utama dan unggulan di Indonesia, serta sebagai pendorong tumbuh dan berkembangnya industri hilir berbasis minyak kelapa sawit. Minyak kelapa sawit dapat diekstraksi dari mesokarp buah kelapa sawit yang mengandung asam lemak jenuh dan tidak jenuh berantai panjang yang tinggi (C:16-0 dan C:18-1),Tujuan dari penelitian ini adalah untuk mengetahui kadar ester paling optimum yang diperoleh dari hasil transesterifikasi crude palm oil dan mengetahui aktivitas antibakteri dari ester hasil transesterifikasi minyak sawit mentah.
Metode penelitian yang dilakukan antara lain, proses transesterifikasi dan pengujian aktivitas antibakteri ester asam lemak. Proses transesterifikasi dilakukan menggunakan variasi konsentrasi katalis basa kalium hidroksida yaitu 1, 2, 3, 4 dan 5 g dengan suhu yang bervariasi yaitu 30°C, 55°C dan 65°C selama 2 jam. Ester yang optimum tersebut dilakukan pengujian aktivitas antibakteri terhadap Staphylococcus aureus dengan metode sumur menggunakan media Nutrient Agar pada konsentrasi 12,5, 25, 50, 100, dan 200 mg/ml.
Berdasarkan hasil penelitian diperoleh kadar ester yang optimum diperoleh dengan jumlah kalium hidroksida 2 g pada suhu kamar yaitu sebesar 99,0571%, dengan jumlah kalium hidroksida 5 g pada suhu 55°C yaitu sebesar 98,4044%, dan dengan jumlah kalium hidroksida 3 g pada suhu 65°C yaitu sebesar 98,5087%. Hasil pengujian aktivitas antibakteri dari ester minyak kelapa sawit yang optimum menunjukan bahwa pada bakteri Staphylococcus aureus memberikan daya hambat yang paling tinggi yaitu 9,7 mm dengan konsentrasi 200 mg/ml.
Proses transesterifikasi dengan jumlah kalium hidroksida yang lebih sedikit dan suhu yang rendah dapat menghasilkan kadar ester optimum senilai 99,0571% dan dapat digunakan sebagai antibakteri dengan konsentrasi 200 mg/ml.

THE ANTIBACTERIAL ACTIVITY TEST THROUGH OPTIMIZED TRANSESTERIFICATION OF OIL PALM
(Elaeis guineensis Jacq)
ABSTRACT
Palm oil (Elaeis guineensis Jacq.) is one of the main agricultural commodities and prime in Indonesia, as well to support the growth and development of downstream industry based on palm oil. Palm oil can be extracted from oil palm mesocarp which contained high of saturated fatty acids and unsaturated long-chain (C: 16-0, and C: 18-1). The purpose of this research was to determine the most optimum level of ester that obtained from transesterification of CPO and determine the antibacterial activity of ester that outcome from transesterification of crude palm oil.
The research methodology included the transesterification process and examined the antibacterial activity of fatty acid ester. The transesterification process done by using variation concentration of alkaline catalyst (potassium hydroxide) amount 1, 2, 3, 4 and 5 g with varying temperatures of 30°C, 55°C and 65°C for 2 hours. The optimum ester was examined its antibacterial activity against Staphylococcus aureus with well methods by using Nutrient Agar media at a concentration of 12.5, 25, 50, 100, and 200 mg/ml.
The results showed that the optimum level of ester which obtained 2 g amount of potassium hydroxide at room temperature is 99.0571%, 5 g amount of potassium hydroxide at 55°C is 98.4044%, and 3 g amount of potassium hydroxide at 65°C is 98.5087%. The result of antibacterial activity in the optimum ester palm oil showed that the Staphylococcus aureus bacteria provides the highest inhibition, that is amount 9.7 mm with a concentration of 200 mg/ml.
The transesterification process with lower of potassium hydroxide and low temperature can produced 99.0571% optimum of ester level and can be used as an antibacterial with a concentration of 200 mg/ml.
