Abstract:

The foxtail millet Waxy gene encodes the granule-bound starch synthase (GBSS), which is the key enzyme in the amylose synthesis. Genomic DNA of the Waxy gene was sequenced in 34 landraces of foxtail millet [Setaria italica (L.) P. Beauv] to clarify the molecular variations at the DNA level among waxy, low amylose or non-waxy genotypes. Foxtail millet is one of the oldest cereals widespread in Europe and Asia of early agriculture era and, interestingly, waxy phenotype has been grown only in specific geographical regions, mainly in continental parts of Southeastern and Eastern Asia, where sticky foods have been favored traditionally and played an important role. Four independent PCR primers, comprising the entire waxy gene, were designed, and PCR products were amplified using the waxy gene-specific primers. DNA sequence was determined using direct sequencing. Many polymorphic sites in the entire waxy gene were detected depending on landraces and their phenotypes. Many of SNPs and indels were shown in the 5¡¯ UTR region amplified. Also, further DNA sequence for the waxy gene will be determined for a total of 150 foxtail millet landraces collected from all over the world.

Furthermore, phylogenetic analysis will be performed to understand the geographical and evolutionary phases for the Waxy gene.
