SUMMARY
Several lines of evidence now support the hypothesis that oestrogens regulate many processes of the male reproductive system under physiological conditions.
This
thesis presents investigations on pubertal developmentin
SouthAustralian Merino
ram lambs and examines the effectof
active and passive immunizations against oestradiol-17ß or oestrone on testicular maturation.Al1 ram lambs were weaned
two
weeks before the startof
theexperiment. They
were kept indoors,in
large pens, under constantlighting (lZL:l2D)
and were fedwith
commercial sheep pellets and lucernehay.
Each experiment on pubertal development started at 10or
14 weeksof
age and lasted sixteen weeks.
A
progressive increase in body weight and testicular volume occurredin
all ram lambs studied.Mean
LH level
tendedto
decrease as the animals pass through pubertywhile no
distinctivevariations in FSH and PRL
secretionswere noticed. Pituitary
responsivessto a
GnRHchallenge
decreasedbetween 14
and30
weeksof
age andprobably reflected
increasing,n¡ç"
^çato|v feedback
by
gonadal steroids as the steroidogenicactivity of
the testisIndeed, mean plasma testosterone
level
aswell
as testosterone secretionfollowing
a hCG challenge increasedmarkedly
asthe
animalsmatured. Total testicular blood
plasmaflow (TTBPF)
when expressed perunit weight of
testis(pl/g/min)
was shownto
decrease as the testis grows, although total bloodflow
per testis(ml/min)
isincreasing.
Subcutaneous scrotal temperature seemed to decrease as testicular size increases. Most of the ram lambs had achieved puberty between22
and 26 weeksof
age as confirmedby
the presenceof
spermatozoain

Detrimental effects have been observedin
some E2-immunized ramlambs. For
instance, we have observed a steep declinein
testicular size towards the endof
the experiment, presenceof
large vacuoleswithin the
seminiferousepithelium and,
in
one lamb, nearly complete absence of germ cells at 30 weeksof
age. TTBPF per testis(ml/min)

weight of testis(pl/g/min)l
was more elevatedin
control thanin
E2-immunizedlambs.
The steroidogenic function of the testis was remarkably enhanced in theE2-immunized
lambs as reflectedby
thehigh
plasma testosterone concentrations, however, body weights were not improved in these lambs (no anabolic effect)'Testicular biopsies have been taken
at22
and 26 weeksof
agein
someof
these ram lambs.This procedure did not seem to affect significantly subsequent testicular development, however, testosterone
secretion
measuredat the end of the experiment (at 30 weeks of
age) wassignificantly reduced in these ram lambs.
In
other experiments, some well-characterized antibodies directed against oestradiol-17ß or oestrone(purified IgG or
complete antiserum, showinghigh specificity
andhigh affinity
invitro )
have been injectedto
intact ram lambs during pubertal development (10ot
14 week- duration) or to oestradiol-implanted castrated ram lambsfor
a short-term period(l
ot 2 weeks).Unfortunately, no difference was found between the control and the passively immunized intact ram lambs, and
only
a slight reduction of the oestradiol effect on gonadotropin secretion have been observedin
the wethers passively immunized againstoestradiol.
This lackof
effect may have been due to an insufficient amount of antibodies injected into the animals (low titre) and/or incomplete neutralization of the hormone (e.g. relativelylow affinity
in vivo ).In conjunction with
thiswork
on puberty, this thesis includes a one year duration experiment using adultMerino
rams actively immunized against testosterone. We have demonstrated that6616F
the persistent increment
in
gonadotropin and testosterone secretions observedir{f}mmunized rams
arenot sufficient to maintain improved
spermatogenicfunction throughout
the year.Furthermore, we have shown that this treatment could lead to a reduction in epididymis weight after twelve months of immunization as
well
as a reduction in the number of mount culminatingin
ejaculation performed during a ten-minuteslibido trial
done on various occasions during the experiment.J
