Additional Professional Activities
During the past years my research emphasis was placed on:
Pyrimidine Metabolism in Bacteria with special focus on the Regulatory Eenzyme, Aspartate Transcarbamolase (ATCase).
All Pyrimidine Enzymes Assays, using different techniques such as Spectrophotometer.
The role of Pyrimidine Enzymes in the regulation of the Pyrimidine pathway and other Chemical pathways.
Genetic organization and biochemistry of prokaryotic DNA.
Practical application of Polymerase Chain Reaction (PCR)
Through these research projects I obtained knowledge and techniques in recombinant DNA technology, amplifying of cloned gene, bacterial strain construction, Making competent cells, transformation, selection and purification of wild type and mutant ATCase by using affinity chromatography. .
The focus of my master and Ph.D research was on pyrimidine metabolism in bacteria with special emphasis on the regulatory enzyme. In my master thesis my research was focused in the study of the regulatory enzyme Aspartate transcarbamoylase (ATCase) in Pseudomonas putida. In this project I dealt with the purification and kinetic characterization of mutant and wild type Pseudomonas putida ATCase. During that time I experienced different techniques such as, strain construction, making competent cells, transformation, selection, and purification of ATCase by using ionic exchange column. In addition to that, techniques in cell biology were also experienced by taking a practical course.
The main focus in my Ph.D research was to study the relation between different mutants in the pyrimdine genes and the production of virulence factors in the opportunistic pathogen, Pseudomonas aeruginosa. This study showed that all mutant strains were deficient in the production of virulence factors. Low level production of pyocyanin, hemolysin, casein protease, and elastase were found in the mutant strains compared to the wild type strain. Swimming, swarming, and twitching motility were also impaired. Many different techniques were experienced during that time including the following:
polymerase chain reaction (PCR) and its different application, hybridization, Southern blot, gel electrophoresis, DNA isolation, Protein purification, and construction of mutant strain.
