ABSTRAK

Atrofi vagina merupakan masalah yang sering dialami oleh wanita menopause yang berkaitan dengan penurunan kadar hormone estrogen. Terapi Sulih Hormon (TSH) dengan estrogen terbukti efektif dalam mengatasi gejala atrofi vagina, tetapi meningkatkan resiko kanker payudara, endotrium dan ovarium. Fitoestrogen merupakan bahan yang berasal dari tanaman, memiliki struktur mirip estrogen, mampu terikat pada reseptor estrogen (ERβ dan ERα), dan dapat menginduksi efek seperti estrogen. Fitoestrogen merupakan alternative yang lebih aman untuk mengatasi atrofi vagina.

Antosianin terbukti memiliki aktivitas fitoestrogen baik invivo maupun invitro. Umbi ubijalar ungu yang terdapat di Bali memiliki kandungan antosianin cukup tinggi. Tujuan penelitian ini adalah untuk mengetahui efek pemberian ekstrak ethanol ubi jalar ungu terhadap ekspresi mRNA ERα dan ERβ jaringan vagina tikus putih yang mengalami ovariektomi. Sebanyak 28 ekor tikus betina yang diovariektomi dibagi dalam 4 kelompok dengan dengan 3 kelompok diberi ekstrak ethanol ubi jalar ungu dengan 3 dosis berbeda (1ml, 2ml dan 4 ml/hari) dan satu kelompok kontrol diberi normal saline 1 ml/hari selama 30 hari. Dilakukan ekstraksi RNA dan qRT-PCR untuk mengetahui ekspresi mRNA gen ERα dan gen ERβ. Hasil penelitian belum bisa dianalisis karena perlakuan terhadap hewan coba masih berlangsung.

ABSTRACT

Vaginal atrophy is a common conditions in post menopausal women associated with decreased of hormone estrogen level. Hormone replacement therapy (HRT) with estrogen proved to be effective in treating the symptoms of vaginal atrophy, but increases the risk of breast cancer, endotrium, and ovaries. Phytoestrogens are substances derived from plants, has a structure similar to estrogen, capable of binding to the estrogen receptor (ERβ and ERα), and can induce estrogen-like effect. Phytoestrogens are a safer alternative to cope with vaginal atrophy. Anthocyanins shown to have activity both in vivo and in vitro phytoestrogens. Purple sweet potato tubers have a fairly high anthocyanin content, so it has the potential to overcome the symptoms of vaginal atrophy. The aim of this study were to investigate the effect of ethanol extract of purple sweet potato on the mRNA expression of ERα and ERβ vaginal tissue of ovariectomized female rats. A total of twenty eight ovariectomized female rats were randomly divided into four groups. Three groups were given ethanol extract of purple sweet potato with three different doses (1ml, 2ml and 4 ml / day) and a control group was given normal saline 1 ml / day for 30 days. After treatment, RNA extracted from vaginal tissue followed by qRT-PCR to determine mRNA expression of ERα and ERβ genes. The results of the study could not be analyzed because the treatment of the animal is still in progress.
