Determining the Threshold Value for Identification of the Goblet Cells in Chicken Small Intestine

seen from the number of goblet cells found in the small intestinal epithelium. The cell identification is required to count the number of goblet cells and in the most cases, identification of goblet cells is carried out through a microscope directly. Although the directly identification saves costs, but the implementation not only takes a lot of time and energy but also is relatively subjective. Here we seek to design a system that can identify goblet cells automatically through segmentation by using two threshold values. The two threshold values are obtained from the gray scale images. Gray scale image is defined as the result of extraction of the hue component in hue saturation value type images. Goblet cells as the main producer of mucin can be colored in the processing of tissue preparation by using alcian blue-periodic acid schiff staining technique. This technique turns out the mucin into blue, so the identification of goblet cells can automatically be conducted by extracting blue features. After that we decide the lower and upper threshold values from the results of extraction which show the object as a goblet cell. In conclusion, we provide the results that the threshold value to separating object from other networks is on the gray scale, with value 176 to 221.

The segmentation using two threshold values proves that the system is able to identify the goblet cells properly.

NTRODUCTION

Chicken productivity is very influenced by intestinal health. Intestinal health can be seen from the number of goblet cells found in the small intestinal epithelium [1].

Goblet cells have a major role in digestion and absorption of nutrients, and also play an important role as the main producer of mucin [2]. Goblet cells are also an indicator of intestines in a healthy or infected condition [3], the more goblet cells found indicates that intestinal in an unhealthy condition [4]. The number of goblet cells can be calculated in direct by using a microscope by identifying goblet cell object one by one. Although direct visual identification has benefits such as low cost and flexibility, but the procedure takes a lot of time and labor [5]. In addition the subjectivity also influences the process and the results are less precise and accurate [6]. Nowadays, automatic research on cell identification has been carried out, i.e image processing on the detection and calculation of the number of red blood cells by performing the initial stages of calculation in the type of image preprocessing to gray scale [7]. At the same time, the method was applied in the identification of the development phase of plasmodium falciparum in red blood cells that preprocessed images into gray scale by extracting the hue component in the color spectrum converted image from RGB (Red, Green, Blue) to HSV (Hue, Saturation, Value).

Thereafter the gray scale image is segmented to separate cells with other networks [8]. But until this time there has

been image processing research yet to identify goblet cells in chicken intestines.

Based on these problems it is necessary to develop a method that can be used to identify chicken intestinal goblet cells automatically by using image processing method. Here the object of goblet cells are separated from other networks through a segmentation process. The segmentation is using multilevel thresholding, in whch there are two threshold values are used to separate goblet cell object with other networks. Some images of intestinal parts that have been prepared with the AB-PAS (Alcian Blue-Periodic Acid Schiff) staining technique [9] are acquired using a microscope which is equipped with a camera and connected to a laptop. The image of the acquisition is in the type of an RGB image, and then converted into an HSV image. The extraction of the hue component is performed on HSV images to obtain a threshold value, so those images with gray scale are generated. The extraction results show that the object of goblet cell is at between two gray scale values.

Then both of these gray scale values are used in the segmentation to identify chicken intestinal goblet cells.

II. RESEARCH METHODOLOGY

A. Goblet Cells

Goblet cells are the main producer of mucin, located in the small intestinal epithelium and have a primary role for digestion and absorption of nutrients [2]. Basically, the number of goblet cells will increase due to the food aflatoxin contamination [4]. Several preparation techniques have been determined for counting goblet cells, one of which is AB- PAS staining technique. The AB-PAS staining technique is a dual-purpose method for detecting goblet cells both neutral and acid mucins [9]. The photomicrographs displaying goblet cells with AB-PAS and mucicarmine are shown in Figure 1. The blue object is a goblet cell, while the remain is another networks.

B. HSV Image (Hue, Saturation, Value)

HSV imagery is a color image in the terms Hue, Saturation and Value. The characteristic of HSV is there are

colors similar to those captured by the human senses. Hue's true colors, such as red, violet, and yellow and are used to determine redness, greenness and so on. Saturation is sometimes called chroma, is the purity or strength of color.

Whilst the value will describe the brightness of color. The value ranges from 0-100%. If the value is 0 then the color will be black, the greater the value, the brighter the colour and new variations emerge from those colors. The HSV model is shown in Figure 3.

RGB to HSV colour conversion is a process of changing from an RGB type image to an HSV type image. RGB to HSV colour conversion is formulated in the following equation:

The component extraction of Hue, Saturation and Value is one of ways that can be used in image processing to determine an object based on colour characteristics.

Component of Hue, Saturation and Value have advantages in red, green and blue spaces in detecting an object [10], with the resulting data class in the form of a double class image.

C. Two Threshold Segmentation

The Two Threshold Segmentation is a method of image segmentation with 2 level thresholds. Here the image is divided into several parts using two threshold values.

Especially, this is implemented if on the histogram there are several peaks that distinguish between one object to another, where the intensity value of object is known to be between T1 and T2. The values searching for the construction can be conducted by looking at the gray scale intensity of the image [11]. The following Figure 2 is an example of a gray scale image that has several peaks on the histogram.

The grayscale object with gray scale values less than T1 and more than T2 turn into black binary images, while the grayscale object with values equal to or between T1 and T2 values change to white binary images.

III. DESIGN AND IMPLEMENTATION

Determination of threshold values was calculated by using Matlab Programming Language. It started with determining the object identified as goblet cells seen in the image until the stage of determining the lower threshold value (T1) and upper threshold value (T2) for further identification of goblet cells. The object identified as a goblet cell was determined by its characteristics including the color and intensity of gray. Gray intensity on the object was randomly selected based on the difference in intensity of the most striking blue differences from 30 RGB images of object in the small intestine broiler tissue part of the jejunum which had been prepared with AB-PAS staining. The steps for determining the threshold values are presented in Figure 4.

The process of determining T1 and T2 values in the image of a goblet cell object includes the following steps:

The first step was selecting the image part that its the object of the goblet cell with the darkest and brightest blue intensity by using the ROI (Region Of Interest)

process of the square function. The image generated from the ROI process was in the form of a RGB- shaped square image. Example of the selection of goblet cell object with the darkest blue and brightest through ROI selection is shown in Figure 5.

The selected image with the ROI process was converted from RGB scale image to HSV. The purpose of this conversion is to select the component hue as a component used in the process of image conversion to gray scale. Example of the RGB to HSV image conversion results shown in Figure 6.

The third step was extracting the Hue components from HSV images. The image generated from the extraction of the Hue component was a gray-faced image with a double class. So in that way, it needed to be converted into uint8 class. If the input value is less than 0 then it is converted to 0, if the input value is more than 1 then it is converted to 255, whereas if the input value is between 0 and 1 then the conversion is multiplied by 255. Example the extraction results of the Hue component are shown in the Figure 7.

The image that has been converted into a gray scale is sought for the highest gray intensity value. Here the

pixel region window provides the value of gray intensity. Gray scale values in regional pixels are in the range of 0 to 255. Then the resulting image and gray intensity values obtained in this last process were stored to find the minimum and maximum gray values of all images of the goblet cells taken. The integrity of the image obtained in this process was 30 images and 30 values of grayness were minimal and 30 values of maximum gray intensity. The display of gray intensity values of grayscale images is shown in Figure 8.

IV. R

ESULTS

Based on the determining process of the threshold value carried out on 30 images, T1 was obtained with a value of 159 and T2 with a value of 221 as presented in Table 4.1.

The minimum and maximum gray intensity values obtained will be used in the two threshold values segmentation process. After that, the results obtained from determining the threshold value are tested to determine the identification accuracy of the threshold values.

The results from testing on 30 images of goblet cell object with the darkest blue intersections and brightest blue intensity were obtained 4 groups of segmentation results. The first group of segmentation results are similar to object, the second group of segmentation results display some object pieces, the third group of segmentation results display two adjacent object, and the fourth group segmentation results are noise and holes.

The first group of segmentation results is similar to the object. In that case it shows that the results of segmentation with two threshold values can separate the goblet cells object with the background be able to be separated. Beside that, the shape is also almost similar to the goblet cell object on the RGB scale. This result is found in 7 images T1 determination and 9 images from T2 determination. The examples of segmentation results with results are similar to the original object shown in Figure 9.

The second group of segmentation results can separate the goblet cell object with its networks. While there are pieces on several sides of the goblet cell object. This result is found in 8 images from T1 determination and 10 images

from T2 determination. The Example results of segmentation with truncated object in some parts are shown in figure 8 and the examples of segmentation results with results similar to the original object shown in Figure 10.

The third group of segmentation results can separate the goblet cells object with their networks even though there are two adjacent objects and sticking together. This result is found in 3 images of T1 determination and 2 images T2 determination. The examples of segmentation results with the results of two adjacent and fused are shown in Figure 11.

The fourth group of segmentation results can separate the goblet cell object with its networks even though there is a noise around and a hole in the object. This result is found in 12 images from T1 determination and 9 images from T2 determination. The examples of segmentation results with results are similar to the original object shown in Figure 12.
