Tables SDC 8: WGA performance assessment. Decreasing numbers of HT29 cells, BRAF V600E heterozygous, were sorted in a 96 well plate: 10, 5, and 2 cells were sorted in 5 different wells (column #1-2, Table S1); 19 single-cell wells were also collected (Table S2).

Sorted cells were subjected to PicoPlex WGA and subsequent amplification/mutation analyses (columns #3-9). HT29 amplified DNA was

subjected to PCR covering 4 different chromosomal regions (columns #3-6) with different length amplicons (located in chromosomes

12p, 5q, 17p, 6q), and to ddPCR for EGFR exon 20, KIT exon 11, BRAF exon 15 (columns #7-9). The detection of the V600E mutation and

the allelic fractional abundance (f.a.) is also reported (columns #10-11). Allelic drop-out/imbalance (ADO/AI) occurrence is indicated in

column #12. nd: not detected; WGA ctrl: HT29 cell DNA subjected to WGA; DNA ctrl:HT29 cell DNA not subjected to WGA.
