It can also measure important parameters of individual RBC with the programmed algorithm such as corpuscular hemoglobin content (CH), corpuscular hemoglobin concentration (CHC) and corpuscular volume (CV). Therefore, we predicted that the fluctuations of the MDS RBCs would be reduced because there should be imperceptible damage to RBC membranes caused by dyserythropoiesis. Using this microscope, we tried to find features or different aspects of MDS RBCs in peripheral blood samples that could be useful to diagnose MDS or to elucidate pathogenesis, little more than the present.

And two samples of Welch's t-test was used for comparison of RBC parameters between MDS patients and healthy individuals. Unlike other anemic features of RBC that we were aware of, such as target cells or increased central pallor area, it was a new cup-shaped morphological peculiarity. Moreover, this microscope can measure important parameters of individual RBC with the programmed algorithm, such as corpuscular hemoglobin content (CH), corpuscular hemoglobin concentration (CHC) and corpuscular volume (CV).

Also, individual RBC parameters can be measured, which are not possible to obtain by the commercial automated hematology analyzer, such as diameter, membrane fluctuation, surface area (SA) and surface index (SI) of an RBC2. Membrane fluctuation is a very exclusive parameter to obtain under the microscope.. continuous and rapid recording of 2-D RI phase images using a high-speed camera. Fluctuation of RBC membranes is acutely affected by various pathophysiological conditions such as diabetes mellitus.

The deformity of RBC is .. determined by various properties such as membrane composition, cytoplasmic viscosity, spectrin network, shape and size4).

Reconstructed 3-D images of RBC

High in SI means that MDS RBCs tend to be more spherical than the normal RBCs. This means that when the RBC volumes are the same, MDS RBCs have less surface area than the normal RBCs, which is ineffective in oxygen delivery. Only pancytopenia with increased MCV and red blood cell distribution width (RDW) was observed in the patient's Wright stained.

The cup shape may be the result of the loss of elasticity after detangling process in the bone marrow. Patients diagnosed with MDS who have chromotropoietic features in the bone marrow should be examined with the ODT setup microscope to obtain 3-D RBC morphology. The cup-shaped RBC has the potential to be the important and only morphological feature found in the peripheral blood for the determination of erythroid lineage dysplasia.

MDS로 진단되고 골수에 영양이상 특징이 있는 환자는 추가 조사를 위해 3차원 적혈구 형태를 얻기 위해 ODT 설정 현미경으로 확인해야 합니다. 골수이형성증후군(MDS)은 병인 및 병태생리학적 기전이 명확하지 않은 클론성 악성 신생물입니다. 또한 자동화된 혈액세포 분석기에서만 측정할 수 있었던 적혈구 혈색소(CH) 함량, 적혈구 혈색소 농도(CHC), 적혈구 용적(CV) 등의 적혈구 지표를 단 하나의 적혈구에서 얻을 수 있습니다. .

또한 적혈구 직경, 막변동, 표면적(SA), 표면지수(SI) 등 자동화된 혈액세포 분석기로 측정할 수 없는 값을 얻을 수 있다. 이에 본 현미경을 이용하여 골수이형성증후군 환자의 말초혈액 적혈구의 특성을 알아보고자 하였다. 서울아산병원에서 검사하고 남은 EDTA 말초혈액 샘플을 사용하였습니다.

실험군은 MDS로 진단된 환자의 검체를 사용하였고, 대조군은 건강검진을 위해 병원을 방문한 환자의 검체를 사용하였다. 또한, MDS 환자를 포함하여 혈구감소증을 동반한 다양한 질병을 앓고 있는 환자의 샘플을 이용하여 적혈구의 3차원 영상을 구현하였습니다. 회절광학현미경은 HT-1S와 HT-1H(Tomocube Inc., Daejeon, South Korea)를 사용하였다.

Tomostudio 소프트웨어(Tomocube, Inc., Daejeon, South Korea)를 사용하여 적혈구의 이미지와 측정값을 얻었습니다. 실험군과 대조군의 적혈구 매개변수 값을 비교하기 위해 상관관계 맵과 2-표본 Welch's t-test를 사용하였다.
