WORKSHOP SUMMARY
Many Bioscience students and professionals find the concepts of enzymology difficult to understand.
The aim of this workshop therefore is to provide an in-depth revision of the basic concepts of enzymology; and to expand these concepts to the actions of enzymes within metabolic pathways. In this workshop we will discuss the implications of the irreversible and reversible Michaelis Menten equation and its implications on enzymatic rates within linear biochemical pathways. We will look at how typical enzyme parameters can be derived from experimental data. Furthermore, we will investigate different reversible inhibitor types and how they affect metabolic pathways. The general concepts of enzymology will be further expanded to 2-substrate reactions and how they can be analysed. We will discuss the function and behaviour of allosteric enzymes and investigate their regulations within a metabolic context. With a thorough understanding of these concepts we will then explore, how enzymes can perform complex functions within a systems biology context. For this we will look at selected systems containing feedback and feed forward loops and analyse the concept of zero- order ultra-sensitivity, resulting in ‘futile cycles’. Finally, we will discuss the allosteric regulation of complex pathways, which lead to emergent behaviour of metabolic pathways. Examples of pathways will be illustrated through systems biology modelling with JDesigner.
