1. Kit qui phan tich da hinh RAPD-PCR Phan ung RAPD-PCR vdi 28 mdi ngiu nhidn thudc cic nhdm OPN, 0PM, UBC, OPO, OPC, S, BiO, OPA da du(?c nhSn thanh cdng. 14 mdi cho da hinh da dupe ding cho phan tich vi udc lu(mg quan he di truyte (bing 2). Cic mdi cdn lai cho cic bang don hinh hoac da hinh khdng r6 rang.
Cic phan doan ADN quan sat thiy trdn gel didn di cd kich thudc nim trong khoang tir 0,24 kb ddn 2,75 kb. Sd phin doan nhan bin ddi vdi moi mdi xe dich til 7 den 22, trung binh 14,8 phin doan/mdi.

EVALUATION OFTHE GENETIC DIVERSITY OF SEVERAL LOCAL TARO (Colocasia esculenta (L) Schott) STRAINS USING RADP MARKERS

Nineteen local taro {Colocasia esculenta) and one Colocasia gigantea strains collected from different parts of the Nortii and North Central region were evaluated for genetic diversity using random amplified polymorphic DNA (RADP) pnmers. Twenty eight pnmers amplified PCR DNA products and fourteen pnraers shown the polymorphisms with an average per cent polymorphism is 86.96, were used to analyze RADPs and disUnguish between accessions. Thirteen pnmers out of these showed high polymoiphic information. Per cent polymorphism varied from 73 to 100 among these polymorphic pnmers. RAPDs showed high geneUc diversity in these accessions. UPGMA cluster analysis of genetic similarity estimates separated the accessions in two main groups at similarity coefijcient of 0,54. All 19 accessions of C.
esculenta divided into four subgroups (I, II, 111, and IV). The results of clustering taro based on the RADP markers is seem too similar to the moiphologically cultivar grouping. On other hand, RADPs were able to distinguish among taro accessions. The high genetic polymorphism and many amplified DNA bands specific to eco-geographical regions founded in the Midland and Northern mountainous areas may iie a scientific basic for in situ conservaUon of taro germplasms in our country. The results showed also that die geneUc improvement of local taro U-ains should be based on molecular variations as well as morphological differences.
