An extracellular phytase (myo-ioositol hexakisphosphate 3-phosphohydrolase, EC 3.1.3.8) bom Asper^llus niger V^\ was purified „'L about 14.7-fold to ^parent liomogeneity with activity recoveiy of 37.4% referred to the phytase activity in die crude extract. ';

PuiificatioD was actiieved by 80-90% ammonium sulphate precipitation m combination with diromatography on ion SP-streamline ' exchange cohnnn, Phenyl-Sepharose hydrophobic cohimn and sephadex G75 gelfiltiation column. The molecular weight of purified ',.

phytase estim^ed by SDS-PAGE in combin^on with gel filtration was a monomeric protein of about 87 kDa. The optimum pH and '^

temperature for phytase activity were at pH 3.5 and SSX, respectively. The enzyme was activated by Ca^ * and inhibited by a number '

of ion including Fe^*, Mg^, Mn^*, Na*, K* and Zn^'' at 1 mM and 5 niM concentrations. iil.
