ABSTRACT

Effect of donor cell sex o n t h e d e v e l o p m e n t of Ban p i g embryos b y somatic cell nuclear transfer indigenous pig breeds of Vietnam are very diverse, now because of spontaneous propagation and poor management, there is a serious decline in number of them. So, establishment a biodiversity conservation system is extremely urgent and necessary. The aim of this study is conservation of Ban pig. In this research, we create Ban pig embryos by cloning method b y using somatic cell line from Ban male a n d female a n d Landrace oocytes In this report, w e present the effect of donor cell sex on the development of Ban pig embryos. Tlie results showed that the cleavage rate, blastocyte rate, hatching rate between two groups were not significant difference However, the number of cells per blastocysts of group using Ban female cell lines was higher than that of group usmg Ban female cell lines (45.1 cells with 37.5 cells). It, therefore, is possible to use Ban female cell Ime as a donor cell to improve the quality of Ban pig embryos by somatic cell nuclear transfer.

Lon ban dia Viet Nam la nguon tai nguyen sinh hoc ra't quy gia voi nhieu tiem nang chua duoc khai thac va phat huy hieu qua. Tuy nhien, hien nay do viec nhan gio'ng tu phat va quan Iy khong chat che, cac gio'ng lon ban dia dang hi Iai tap nhieu, ma't dan di nhirng dac tinh quy. Tren the gioi, nhan ban vo tinh tren lpn dupc thuc hien thanh cong tir nhiing nam 1989 trong nghien ciiu cita Prather va ct\', ong da sir dung te'bao hen nhan va te'bao ciia phoi de cay vao tning. Da tao ra dugc Ion con bang phuang phap nhan ban vo hnh (Boquest va ctv, 2002), nhung su khac nhau ve hieu qua tao phoi nhan ban vo tinh due va cai van chua dupc chiing minh ro rang. Kumore va ctv, 2008 cho bie't, hieu qua san xua't Ion nhan ban vo tinh sii dung nguyen hao spi thai nhi dugc thu tir thai nhi dire va thai rJii cai co su dao dgng trong pham vi 0,64-0,90%.

O Viet Nam, Biii Xuan Nguyen va ctv (2006) da tao phoi Ign Ban nhan ban vo tinh tir cac te'bao sinh du5ng thu tir mot con Ipn Ban trudng thanh. Ket qua cho tha'y ty Ie phan chia dat 81,2% va ty le phat tnen den giai doan phoi nang la 12,5%. Viec tao phoi sit dung trung va tinh triing dong lanh da dupc mpt so tac gia tieh hanh tren giong Ipn Ban, cac nghien cuu da dugc cong bd' nhu nang cao kha nang thu tinh o'ng nghiem sir dung tinh trimg Ion Ban ddng lanh (Nguyen Viet Linh va ctv, 2018), tao phdi til' te bao tning Ion Ban ddng lanh (Somfai va ctv 2019).

Nham muc dich bao tdn nguon gen ciia cac gid'ng lcm ban dia Viet Nam, tao hen de cho cac nghien cuu tie'p theo, chiing tdi tieh hanh nghien ciiu:"Anli hudng cita hai ddng te bao due va cai dugc phan lap tir manh tai Ign de'n Iiieu qua tao phdi nhan ban vd tinh tren Ipn Ban". Nghien ciiu nay ciia chting tdi la budc di tie'p theo hen chang dudng nghien ciiu va phat trien cdng nghe sinh hoc sinh san de bao ton va phat tnen cac gid'ng Ign ban dia Viet Nam.

2. VAT LIEU VA PHIMNG PHAP 2.1. Vat lieu

Hda chat sir dung trong thi nghiem dupc mua tii' hang Sigma-aldrich. Buong triing Ipn Landrace va te bao smh dudng Ipn Ban due va cai dugc thu tiJ manh tai. Thi nghiem duoc thu'c hien hr thang 3/2019 deh thang 11/2019.

2.2. Bo tri thi nghiem

2.2.1. Phuang phdp thu vd nuoi thanh thuc trung lon

Buong triing Ign Landrace dugc thu ba- ld md va chuyen ve phdng thi nghiem trong vdng 1 gid trong nudc mud'i sinli Iy d SS-S/^C.

Buong triing sau dd dugc nia bSng dung dich PBS cd chira them khang sinh. Dich nang hiing dugc hiit tir cac nang tning nam tren be mat buong h u n g cd kich thudc 3-6mm bSng dng tiem va bom vao cac o'ng falcon 15ml dat d 37''C. Sau do cac te bao tning duoc chuyen vao mdi trudng khdng cd hormone va dibutyryl cAMP va nudi trong 22-24 gid tie'p theo, cac te' bao tning dupc nudi d dia 4 gie'ng chiia 500[JI mdi trudng nudi dugc bao phii bdi dau khoang va dupc dua vao hi nudi vd trimg chiia 5% CO, d 38,5"C, do am bao hda. Mdi gie'ng nudi khoang 50 te'bao tning,

bao se dupc kiem tra td'c dd tang h u d n g thdng qua viec de'm so lugng te bao bang each sii dung buong de'm hong cau (hemocytometer) tai thdi diem trudc va sau khi cay chuyen.

Trudc khi chuyen nhan 3 ngay, te bao dugc giai ddng va nudi trong DMEM 10%. Te'bao su dung cho chuyen nhan te' bao sinh duang la nhiing te'bao d cac Ian cay chuyen tir 2-6.

2.2.3. Phuang phdp chuyen nhdn te'hdo sinh duong lan

Chuyen nhan te bao sinh dudng duac thuc hien tren he thd'ng vi phau thuat voi may Piezo theo Otoi va ctv (2016). Theo do, te bao triing sau khi nudi thanh thuc se tie'n hanh Ioai bd sach lap te bao hat (cumulus) trong men hyaluronidase 10%. Triing thanh thuc vdi the cue rd rang dupc loai bd the cue va nhan trong mdi trudng TCM 199+10% FBS bang he thd'ng kim vi phau thuat Te'bao sinh dudng lgn don le duoc ca'y vao trung tao phuc hop trung - te'bao. MEM Non-Essential Amino Acid Solution (Sigma). Sau 5 ngay nudi bo sung them 10%

FBS. De xac dinh trang thai va cha't luang, phdi d ngay thii 7 dugc rira 3 Ian qua PBS cd chiia Img/ml polyvinylpyrrolidone (PVP).

Co' dinh phdi hong gipt lOOpl dung dich paraformaldehyde 4% trong PBS, pH 7.4 hong 1 gid d nhiet dp phdng. Rira Iai phdi 3 Ian trong PBS/PVP

Thi nghiem 1: So sanh ke't qua phan lap va nuoi ca'y te' bao sinh dudng Ion Ban due va cai trong mdi trudng Dulbecco's Modified Eagle's (DMEM) cd bo sung 10% Fetal Bovine Serum(FBS) qua cac Ian ca'y chuyen.

Thi nghiem 2: Nudi thanh thuc hiing lon Landrace lam nguyen lieu cho nhan ban vd tinh.

Thi nghiem 3: So sanh chat luong phdi lan Ban nhan ban vd tinh su dung 2 ddng te bao sinh dudng dupc phan lap d tren. Thi nghiem dirge thuc hien vdi 5 Ian lap lai.

2.3. Xu ly so lieu

So Heu dupc trinh bay dudi dang trung binh {Mean)±SE. Cac sd' lieu sau khi chuyen sang dang arcsin dugc xir Iy bang cdng thii'c ANOVA va so sanh Tukey's. Cac so'lieu duac coi la cd y nghia thdng ke khi P<0,05.

3. KET QUAVA THAO LUAN

3.1. Phan lap va nuoi cay te bao sinh duong lpn Ban due va cai

Te'bao sau khi nudi ca'y so cap, dupc ca'y chuyen vdi mat dp 10^ te'bao/gie'ng trong dia 4 gie'ng. So'te'bao tang sinh dupc ghi nhan sau 4 Ian ca'y chuyen lien tie'p vdi 3 ngay nuoi ca'y.

Yoo va ctv (2017) tie'n hanh theo doi anh hudng cua gidi tinh ddng te' bao den su phat trien aia phdi in vitro va in vivo cho tha'y ty Ie phdi nang trong tao phdi nhan ban vd tinh in vitro (d te'bao Ion cai la 11,9% va lgn due la 11,3%), cao hon so vdi in vivo (d te'bao Ign cai la 7,2% va lgn due la 10,6%). Nhu vay, ty 1| hinh thanh phdi nang sir dung te' bao Ign d^c va te'bao Ign cai trong nghien cuu ciia Yoo va ctv (2017) tuong duong vdi ket qua trong nghien ciiu cua chiing tdi.

Nghien cuu trudc day cho bie't ty le chet bat thudng ciia Ipn con sau sinh dupc tao ra tir ddng te bao sinh dudng Ipn due cao hon so vdi te' bao sinh dudng lgn cai (Yoo va ctv, 2017). Trong so cac Ign con dupc nhan ban da chet trong vong hai tuan sau khi sinh, ty le chet d nhdm sir dung ddng te' bao vd tinh due eao hon so vdi ddng vd tinh cai (41,2 so vdi 33,3%). Cac dii lieu thu dupc tir cac nghien aiu tnrdc do thi kha nang tao phdi nhan ban tir te' bao sinh duong cd the do sir khac biet cua eac ddng te'bao eho. Cu the^ su ba't thudng ve kieu hinh cua Ign nhan ban cd the do tac dpng ciia qua trinh nudi cay in vitro cua cac te'bao cho. Tuy nhien, ddng te'bao due cd gay chet sdm cho lgn con hay khdng thi can phai nghien cuu them. Cho deh nay, chua cd ldi giai thich nao cho su khac biet ve nguyen nhan gay che't cua Ipn con dugc nhan ban tir hai ddng te' bao due va eai, do do eac nghien ciiu tie'p theo la can thie't de lam rd van de ciing nhu dua ra bien phap ky thuat Iam tang ty le sd'ng sdt ciia lpn con nhan ban vd tinh.

4. KET LUAN

Ty Ie tao phoi nhan ban dupc tao ra tir te' bao lgn due va cai la khdng cd su khac biet, nhung tir cac chi tieu theo ddi cho thay cha't luong phdi cua ddng te'bao lgn cai td't hon so vdi dong te' bao I^n due. Nhu vay, gidi tinh ddng te'bao cd anh hudng nhat dinh de'n kha nang phat trien ciia phdi lgn Ban nhan ban vd tinh. De nang cao ke't qua nhan ban vd tinh, cd the sir dung ddng te'bao cua con cai lam te'bao cho. Ket qua eiia nghien ciiu gdp phan cho viec sii' dung ddng te'bao cua nhiing gid'ng lgn ban dia Viet Nam nham muc dich bao ton da dang sinh hgc.

L6 1: de d u a c chan tha t v do, khong bo sung t h u c an tmh; 16 2 d e d u o c chan tha t u do, bo sung 300g ngo nghien/con/ngay; 16 3 d e d u o c nuoi nh6't, cho an tu d o t h u c an xanh, bo sung 300g thii'c an hon hop/con/ngay. Ke't qua nghien cuu cho thay khoi l u g n g ciia de liic ke't thuc thi nghiem 616 1 la 16,78kg, 16 2 la 17,63kg; 16 3 la 20,08kg. Sinh t m o n g tuyet doi lan lugt la 47,59; 56,30 va 84,44 g/con/ngay; sinh truong t u o n g doi la 29,34; 33,77 va 46,88%. Hieu qua kinh te'cua cac 16 tii' cao xuong tha'p lan luot la 16 3 lai 3.603.483VND, sau d o la 16 1 lai 1.168.533VND va thap nhat la 16 2 iai 746.133VND, Ket qua so sanh thong ke da p h a n cac chi tieu o i a 16 3 deu sai khac co y nghia thong ke voi 16 1 va 16 2 {P<0,05), n h u n g giiia 16 1 va 16 2 kh6ng sai khac nhau co y nghia thong ke (P>0,05).

T h e study of the effect of raising methods on the growth performance and growth profit margin of goat production at Thainguyen

The study of the effect of raising methods on the growth performance and growth profit margin of goat production was conducted at the Goat Breeding Model of the Faculty of Animal Science and Veterinary Medicine - Thai Nguyen University of Agriculture a n d Forestry in order to prove the its effechveness to economic results for farmers. A total of 18 male goats at 6 m o n t h s of age h a d beginnmg of the experiment of Lot 1,2 and 3 were 12,50,12.58 and 12 48kg (P>0,05) divided into 3 lots' Lot 1 was free-grazed, without supplemented concentrate; Lot 2 was grazed freely, supplemented with 300g of crushed com per head when returning to the stable.
