SUMMARY

Recombinant proteins produced from Gram negative bactena (E coli) is often contaminated widi endotoxin during extracti(««S proiem punfication. Due to dieir high toxicity in vivo and in vitro, endotoxin removal &om injection products is essential. In 4"

study, we developed a method to remove endotoxin in recombinant IL-2 products using reverse phase chromatograihy RP-tSIf combmmg with ultrafiltration widi nulipore membrane of 30 kDa treated with 0.1 M CaClj. Endotoxin concentration was detemuwd by Limulus Amcl)ocyteLysate (LAL) kit measured at 405-410 nm in spectrophotometry. Results showed that >94% endoloxinwis, removed from IL-2 products usmg ultiafilier treated withO.l M CaClj. Usmg diis mettiod togettier widi RP-HPLC endotoxin«
