Bdnh gut (Thong phong) la bdnh roi loan chuyen hda acid uric (AU), lam tdng acid uric mau, ling dong tinh the natri urat trong cdc to ehdc nhu- mang hoat djch khdp, syn xu-ong, gan, to ehdc du-di da. Ddc tru-ng cua bdnh la nhdng dot vidm khdp cip, tai phdt gdy dau dd ddi. Lau dai bdnh trd thdnh vidm khdp bien dang man tinh. Thuoc lam giam du-oc nong do acid uric mau se cd tdc dyng dieu tri bdnh gut.

Thudc tan du-oc dung dieu trj bdnh gut cdn rit it, gia thanh cao va cd nhieu tac dyng khdng mong muon nhu- gay dau byng, tidu chay, soi than, de chd tuy xu-ong, vidm day thin kinh, ddc vdi gan, ddng mau rai rac... Trong khi dd dan gian cd nhiiu bai thudc dung dieu trj gut cd hidu qua ma cd it tac dyng khdng mong muon. Vi vay vide nghidn cdu de ap dyng cac bai thuoc ddng y vao didu tri gut la c i n thiet. Qua chudi hot {Musa brachycarpa Back. - ho Chudi Musaceaej.

Cu ray {Alocasia macrorrhiza (L.) Schott, ho Ray Araeeae).

Ca 2 du-oc lidu du-oc thu hdi tai xa NghTa Phu huydn NghTa Hu-ng tinh Nam Djnh vdo thang 9 ndm 2008. Nguydn lidu sau khi thu hdi, rda sach, thdi Idt mong, phoi khd, sao vang, trdn theo ti Id:

chuoi hdt: 30g, cu rdy: 50g. roi dem sic '^' Nu-dc sic du-oc cd thdnh cao dac den ty Id 4:1. Khi cho chudt uong, hda cao nay vdi nu-dc i m tao thdnh cao Idng theo ti Id thich hop. Lieu dung trong thi nghidm Id lieu tinh theo du-oc lieu khd.

Dong vat nghien CLFU

Chudt nhit tring chung Swiss, khde mgnh, ca hai giong, trong lu-ong td 24 - 26g, do Hoc vidn Quan y cip. Dong vat sau khi mua v i du-oc nudi 3 ngay tai chuong nudi sue vat cua Tru-dng dai hoc Du-oc Ha Ndi tru-dc khi thi nghidm.

Chudt du-oc an vidn thdc an chuan danh cho chudt va du-oc udng nu-dc ty do.

Phu'O'ng phap nghien ciru

Tdc dung ha acid uric mau tren chugt binh thwang

Chudt nhit du-oc chia ngiu nhidn thanh 5 Id, mdi Id 8 eon.

Ld chdng: Uong nu-dc mudi sinh ly.

Ld thd 1: Uong cao long bai thudc chuoi - ray, lieu 8g/kg chudt nhit.

Ld thd 2: Uong cao long bdi thudc chudi - ray, lieu 16g/kg chudt nhit.

Ld thd 3: Uong cao long bdi thuoc chudi - rdy, lieu 32g/kg chudt n h i t

ChuOt d cac Id du-oc uong nydc mudi sinh ly hode thuoc nghidn cdu vdi t h i tich 0,2ml/10g chudt trong 5 ngay liin. Ngdy thd nam sau khi cho uong thuoc 2 gid thr tien hdnh iiy mau va xdc dinh nong do AU mdu '^'^'

Chudt nhit dyoc chia ngiu nhidn thanh 5 Id, mdi Id 15 con.

Ld chdng b$nh: Uong nydc mu6i sinh ly.

Ld doi chiiu: U6ng allopurinol liiu lOmg/kg chudt nhit.

Ld thd 1. U6ng cao long bdi thuie chuii - rdy lieu 8g/kg chudt nhit.

Ld thd 2: U6ng cao Idng bdi thuie chuii - ray lieu 16g/kg chudt nhit.

Ld thd 3 U6ng cao long bdi thuic chuii - ray lieu 32g/kg chudt nhit.

Chudt d cdc Id du-oc uong nu-dc muoi sinh ly hode thuoc nghidn cdu vdi thi tich 0,2ml/10g chudt trong 5 ngdy lidn tiip. Ngdy thd ndm gdy tSng AU mdu bing cdch tidm mdng byng kali oxonat liiu 300mg/kg 1 gid tnj-dc khi cho u6ng thuic lin eu6i.

Sau khi tidm kali oxonat 3 gid thi tiin hdnh l^y mdu vd xdc dinh nong dd acid uric trong huyit tu-ong '^•^'

Xdc dinh nong dd AU trong huyit tu-ong chuOt bing phu-ong phdp enzym: Acid uric bj chuyin thdnh allatoin vd hydroperoxid nhd enzyme uncase.

Hydroperoxid phan dng vdi 4-amino antipyrin vd 3,

5-diehloro-2- hydroxybenzen sulfonic acid (DHBS) tgo thdnh ehromogen, chit ndy dyoc djnh lu-p-ng bing phyong phdp do quang d bydc sdng 520 nm.

Ty 1$ giim ning dd AU trong huyet tyong cua Id thd so vdi Id chdng du-oc xdc dinh dya vdo cdng thdc (1).

Tren chudt binh thu-dng bdi thuic chuii - rdy vdi 3 lieu sd dyng trong nghidn cdu, liiu 16g/kg chudt nhit tring cd tdc dyng giam ning dO AU manh nhit, giam 25,32% so vdi Id chdng vd cd y nghTa thing kd (p < 0,01). Liiu 8g/kg chudt nhit tring Idm giam ning dO AU mdu 11,7% so

vdi Id chdng (P>0,05). Tuy nhidn, so s^nh gida liiu 16g/kg vdi liiu 8g/kg thi khdc nhau khdng cd y nghTa thing kd (p > 0,05). Liiu 32g/kg lam giam ning dO AU mdu khdng ddng k i ( p>0,05)

Qua qud trinh nghidn cdu tdc dyng ha AU mau cua bai thuoc chuoi - rdy trdn chudt thyc nghidm chung tdi rut ra mdt so k i t ludn nhu- sau:

Trdn chudt binh thu-dng: l i i u 16g du-oc lieu/kg trdn chudt n h i t t r i n g cd tdc dyng Idm giam ndng do AU mdu 25,32% so vdi Id chdng (p < 0,01). Hai lidu 8g/kg vd 32g/kg chudt n h i t tring giam khdng cd y nghTa thong kd so vdi Id chdng (11,7% va 0,65%)

Trdn chudt gdy tdng AU mdu b i n g kalioxonat 300mg/kg: bdi thuoc chuot rdy lieu 8g/kg vd 16g/kg chudt n h i t t r i n g cd tdc dyng lam giam nong dd AU mdu tu-ong dng Id 25,70% vd 20,57% so vdi Id chdng (p < 0,05).

S u m m a r y

On normal mice, aqueous extracts from Musa balbisiana and Alocasia macrorrhizos at

the dose of 16g/kg reduced the serum uric acid level by 25.32% as compared to the control (p<0.01). Whereas, in mice with potassium oxonate-induced hyperuricemia, aqueous extracts from Musa balbisiana and Alocasia macrorrhizos at dose of 8g/kg and 16g/kg reduced the serum uric acid levels by 25.7%

and 20.6% as compared to the control (p<0.05).
