Background: Stem cells are cells specialized cell, capable of renewing themselves through cell division and can differentiate into multi-lineage cells. Adipose-derived mesenchymal stem cells (ASCs) display immunosuppressive properties, immunomodulatory features, secrete cytokines and immune-receptors which regulate the microenvironment in the host tissue.
Multilineage potential, immunomodulation and secretion of anti-inflammatory molecules makes MSCs an effective tool in the treatment of chronic diseases. Objectives: The aim of this study was to investigate the concentration and the incubation time enzyme collagenase. Materials and Methods: research animal model mouse Swiss. In the present study, we have explored the concentration and incubation time enzyme collagenase for isolate ASCs and culture MSCs derived from adipose tissue ASCs. Results: 1 mgfats / ml collagenase. incubate for 50 minutes, we have
the highest monolayers: (4.77± 0.31)xl6^ cells / g and 69.33% cell ^owth efficiency.
Conclusions: Using enzyme collagenase. incubated for 50 minutes, we received the hightest mesenchymal stem cells.

II. PHU'ONG PHAP NGHIEN CUtJ

50 phiit ttong tii dm d nhiet dp 37°C, khuay nhe liSn tuc sau 10 phiit. Lpc bdng mang lpc 70pm va rita sach vdi ImL PBS. Dich dugc Igc cho vao dng eppendorf, ly tam 2500 vdng/phiit ttong 5 phut d 37°. Loai bd dich ndi, cho them 1 mL PBS, ldc dlu, ly tam ldn 2, 2500 vdng/phiit ttong 3 phiit d 3TC. Loai bd dich ndi va cho vao 0,5 mL DMEM/F12, 10% FBS, 1% antibiotic/antimitotic, 1% L-glutamine va 1% penicillin/sti^ptomicin [12],

3.1. Ket qua phan l^p va nudi cay te b^o v61 cac nong dd collagenase khac nhau TiSn hanh thu man 5 dgt, chugt cd khdi lugng trung binh la 56,23 ± 0,73 g. Chiing tdi triSn khai hai Id thi nghiSm vdi ndng dp collagenaselmg/mL vd collagenasel,5 mg/mL Sau loc, ly tam, dim sd lugng t l bao vdi Trypan Blue (Hinh 3.1), sd lugng te bao chet (bdt mau xanh tim) la rat it, cho thay collagenase hieu qua cho viec phan lap tS bao don nhan tir md.
KSt qua phan \^p tS bao don nhan cd su khac nhau giiia hai ndng dd collagenase 1 mg/mL va 1,5 mg/mL. Sd lugng t l bdo don nhan thu dugc ty IS thuan vdi nong dp collagenase. So te bao dan nhdn thu dugc d ndng do collagenase Img/mL: 4,24 xlO^ ± 0,33 xlO^ t l bdo/g va d ndng do collagenase 1,5 mg/mL: 5,22 xlO^ ± 0,52 xlO^ t l bao/g.
Tuy nhiSn d nong dp collagenase 1,5 mg/mL mac dii phan lap dugc sd lugng te bao don nhan nhiSu ban nhung van cd mdt vai t l bao chit. Trong khi dd, d ndng d^ collagenase 1 mg/mL, phan lap dugc sd lugng t l bao don nhan it hon nhung hdu nhu khdng cd t l bao chit (Hinh 3.1).

Tiin hanh thu mdu 5 dgt, chugt cd khdi lugng tirung binh la 55,15 ± 0,71 g. Cdt nho 1 g md mo, cho vdo 1 mL DMEM/F12 cd collagenase ndng dd 1 mg/mL, hai nghiSm thiic vdi hai tiidi gian u khdc nhau dugc thuc hien:

Tir cac kSt qua thu dugc chiing tdi nhan thdy, cac mo md ttdng dudi da la nguon khai thac tS bao trung md cd thS thay cho ngudn dich tuy xuong, ddy rdn. Phan lap te bdo don nhan vai ndng do enzyme collagenase 1 mg/mL va li 50 phiit cho kit qua cao. Tit dd, chiing tdi tiSp tuc nghiSn cim, ddnh gid cdc dac diim cua t l bao ttong qua trinh nudi cdy vd biet hda in vitro cua MSCs.

3.3.1. Ket qua nuoi c^y so cap
Cac te bao don nhan sau khi thu nh$n duac nuoi cay trong moi truong DMEM/F12,10% FBS, 1% ABAM, 1% L-glutamine va 1% penicillin/streptomicin.
Sau 24 gio, cac te bao bam. Sau 72 gio, t6 bao trai, co hinh dang hinh thoi dac trung. Sau 5-7 ng4y nuoi ciy, te b4o hpp dong, trai rong va tang sinh. Sau 10-15 ngay nu6i cay, m|t do tS bao dat 70-80%, tiSn hanh cdy chuyk.

So vdi nhiSu nghiSn cim trudc ddy nhu nghiSn cim cua Yang (2011) khi tach tii md ngudi [14], Niyaz (2012) khi tdch tir md chugt [13], Mohammadi-Sangcheshmeh (2013) khi tdch tir ma lac dd [12], thdi gian tl bdo bam va ttai cua chiing tdi cham ban.
Sau 72 gid quan sdt trSn kinh hien vi, cac te bao ttai vdi hinh thai dac tnmg (gidng nguyen bao sgi), nhu vay te bao chiing tdi thu dugc sau nudi cay la te bao tnmg md.
3.3.2. Ket qua cay chuyen tang sinh
Khi bl mat binh nudi khdng cdn dien tich dl cdc tS bao phat triln, cay chuyin de cung cap khong gian cho tS bao phat trien. Sau 24 gid, tS bao bat dau bam dinh va trai dang hinh thoi dac trung va tang sinh. Sau 4-5 ngay, te bao hgp ddng, trai dSu trSn bS mat binh nudi va tiep tuc tang sinh manh.

Sau 24 gid khi tl bao da bam, cdn bd sung chdt cdm iing biet hda. Theo doi tir 24 gid din 2 tudn, nhan thay cd sy: thay d6i hinh dang ciia tS bao khi nudi ttong mdi trudng biet hda: tl bao tir dang thoi dai ddn ddn co ttdn lai. Sau 4-5 ngay, bdt ddu xudt hien cac tS

Khi so sdnh vdi tS bao ddi chirng (nudi ttong mdi trudng DMEM/F12, 10% FBS vd khdng cd chdt bam img biet hda), cdc tS bdo khdng thay ddi hinh dang vd khdng bdt mau thudc nhudm Oil Red-O (Hmh 3.7).

IV. K E T LUAN
Tir nhitiig kSt qud nghien ciiu tten chung tdi nit ra ket luan:
- 6 nong dp 1 mg/mL ttong 50 phiit, sd tS bao don nhan thu dugc: 4,77x10 tS bao/g va sd lugng te bao phat triln dat 69,33%. p a y la kit qua tdt nhdt.
- O ndng dd collagenase 1,5 mg/mL, sd te bao don nhan thu dugc: 5,22x10 te bao/g va sd lugng t l bao phat triln d^t 54,42% cho thdy ndng do collagenase cao da lam anh hudng din khd ndng hdi phuc t l bao, dan din giam kha nang phdt ttiln ttong qua tiinh nudi cdy in vitro.
- Nudi cdy so cdp: t l bdo don nhan bdt ddu bam sau 24 gid, ttdi sau 48 gid. Sau 10-15 ngay nudi cay deit mat do cdy chuySn (70-80%), cd d ^ g hinh Ihoi dac trung.
- Nudi cay thii cap: te bao bam va ttai dang hinh thoi sau 24 gid nudi cay va tang sinh manh, sd lugng t l bao phat tiiln dgt 69,33 ± 1,49%.
- Biet hda tS bao thanh t l bao tao md: Cac t l bao tao mo xudt hien sau 4-5 ngay khi nudi ttong mdi trudng cd chat cam img biet hda. Cac t l bdo nay bdt mau dd khi nhugm vdi thudc nhupm Oil Red-O d ngay thii 10. Cac k i t qua nay da khdng dinh cac te bao thu dugc la t l bao goc trung md.
