2 . PHUONG T r a VA PHUDNG PHAP 2.1. Phirong t i ^

Mdi tnrdng duoc su dung de phan lap vi khuan:

Burk's, NBRIP. Trong dd, thanh phdn cua mdi hirdng Burk's (g L^ gom: 10 sucroza, 0,41 KHaPO^, 0,52 K2HPO4, 0,05 NajSO^, 0,2 CaCl^, 0,1 MgS04-7H20, 0,005 FeS04-7H20, 0,0025 Na2Mo04*2H20 va 20 aga. Tuong ta, thanh phan cua mdi hirong NBRIP (g L') bao gom: 10 glucoza, 5 Ca3(PO,)2, 5 MgCl.-eHaO, 0,25 MgS04-7H20, 0,2 KCl, 0,1 (NH4)2S04 va 20 aga.

2.2. Phuong phap 2.2.1. Phan lap vl khuin

30 mSu dat viing rd cay liia miia ndi duoc thu thap tai xa Vinh Phuoc, huyen Tri Tdn, tinh An Giang. Sau dd, can 1 g mau dat va 99 ml nude cat da dugc khi> tning cho vao binh tam giac va duoc dat frdn may Mc frong 12 gid vdi tdc do 200 vdng/phiit.

Dung dich duoc Idng frong 3 gid, hiit 0,1 ml dung dich nude frong frai ddu fren dia pefri da chuan bi san mdi trudng Burk's khdng N (de phan lap cac ddng vi khuan cd dmh dam), mdi frirong NBRIP chua P khd tan (dd phan lap cac ddng vl khuan hda tan P), d^ khd va u d nhiet dp 30*C. Sau 24 - 48 gid cac khuan lac xu^t hien h-en be mat mdi trudng duoc hdp tuc cay sang dia kliac cho den khi cac khuan lac thuan. Cac khuan lac xuat hien fren mdi trudng Burk's dupc cay sang mdi h-udng NBRIP. Cac khuan lac nay dupc sii dung cho viec danh gia kha nang hda tan P.

2.2.2. Md ta dac diem khuan lac

Md ta hmh thai khuan lac gdm mau sac, hinh dang, dang bia khuan lac va dp noi. Md ta mpt sd d$c diem te hao gom hinh dang va kha nang chuydn dpng ciia khuan lac. Nhudm Gram vi khuan d^ xac dinh gram vi khuan.

2.2.3. Danh gia khi nang hda tan P ciia cac khuan lac diroc phan lap

xac dinh lupng P hoa tan bdi vi khuan frong mdi h-udng NBRIP chua P khd tan. Vi khuan sau khi nudi frong mdi trudng long 48 gid dupc su dung dd danh gia kha nang hda tan P. Dimg 0,5 ml dung dich cua mdi khuan lac cd 0 0 ^ , , la 0,5 cho vao dng nghidm chua san 5 ml mdi tnidng NBRIP long va lac vdi tdc dd 120 vdng/phiit frong 15 ngay. Dung dich ddi chimg la mdi frirdng NBRIP nhung khdng cd sii dung vi khuan. Luong P hda tan b cac thdi diem dupc xac dinh bang phuong phap so mau frdn may quang phd a buoc sdng 880 nm.

3.1.1. Phan lap vi khuan vimg rd cay lua mua ndi

Ket qua cho thay tdng sd 53 ddng vi khudn duoc phan lap tir viing rd liia miia ndi fren hai loai mdi trudng Burk's va NBRIP. Trong dd, cd 40 dong vi khuan duoc phan lap frdn mdi trudng Burk's va 13 ddng vi khuan diroc phan lap fren mdi trudng NBRIP.

The floating nce is conserving in An Giang that associated wilh the maintenance of the community of microorganisms. This research was conducted to select the potenUal rhizosphenc bacteria from acid sulfate soil cultivating of floating nce possessing the ability of phosphorus solubilization. The resulls showed that 53 colonies were isolated rhizosphenc soil from Burk's and NBRIP media. Of those. 5 colonies producing the highest phosphonis concentration were selected as strains 22-BD6. 23-BD6, 27- BD6a. 24-BD6C and lO-BD6b 14.15 - 16.51 mg P,Os L' from Burk's medium. Besides. Uie capacity of P solubiiizing of shains 28-ND6b and 3-ND6 isolated from NBRIP medium were signiiicanUy higher than odier colonies wiUl 6,33 - 7.13 mg PjOs L' after incubahng 15 days. It should be idenUfied die selected bactena and apply under nethouse or field to reduce the volume of phosphorus for plant.
