•t 1,3% va 26 ± 0,12%, tuong ling d cong thirc thi nghiem soc muoi 1,5% va 2,5% sau khi chuyen sang pha hai 10 ngay.

MO' BAU

Astaxanthin (3, 3'- dihydroxy P) la dan xuat ciia p-carotcnoid dugc sir dung phd bien trong nuoi trdng thiiy san (thuc an bd sung cho ca hoi, ca canh), cong nghiep Ihiic phara (chat raau ty nhicn), dugc pham (chat chdng gxi hda, tang cudng klia nang mien djch, phdng chdng ung thu) va thyc pham chirc nang [13, 17].

PHUONG PHAP NGHIEN ClTU Vat lieu

Chung vi tao H. pluvialis Flotow phan lap tai tinh Hda Binh (nam 2009) do phdng Cdng nghe tao, Vien Cdng nghe sitih hgc cung cap.

Tao dugc luu gift va nhan gidng so cap trong mdi trudng C, d 25°C, cudng dp chieu sang 1,5 klux vdi chu ky sang tdi la 12: 12 gid, Thanh phSn mdi trudng C va RM theo cdng bd ciia Dang Diem Hdng va nnk. (2010) [10].

Phifong phap

Tao H. pluvialis dugc nudi trong mdi trudng C, dudi cac dieu kien tdi uu cho sinh trudng (25°C, cudng dp anh sang 1,5 klux, chu ky sang' tdi la 12: 12 gid). Sau 5 ngay nudi (cac te bao chii yeu d trang thai sinh dudng, chiem 80- 90% tdng sd te bao), djch nudi cay dugc ly tam d 6000 vdng/5 phiit. Loai bd phan djch tren, thu can te bao va bd sung mdi trudng RM cd chira cac nong dd mudi khac nhau de tien hanh thi nghiem,

Nghien cihi anh hudng ciia ndng dg mudi len sinh trudng ciia tag H. pluvialis: Thi nghiem dugc tien hanh trong radi truong RM d binli tam giac 250 mL vdi ndng dp NaCl khac nhau: 0%;

0,2%; 0,4%; 0,6% va 0,8%. Mat dp te bao tao ban dau trong cac cdng thite thi nghiem la 6 >=

10'' te bao (TB)/ml, Tao dugc nudi d cac dieu kien tdi uu cho sinh trudng nhu md ta d tren.

Sau dd, dich tao nudi trong pha I se dugc ly tam va thu te bao. Can te bao dugc hda trong mdi trudng RM mdi cd bd sung NaCl d ndng do 0,8; 1,5 va 2,5% (pha n). Lac deu va chia djch tao vao cac binh tam giac 500 va 1000 ml tuong img vdi 350 va 500 ml djch tao/binh. Cac binh nudi tao dugc dat trong dieu kien nhu md ta d tren. Pha 11 trong thi nghiem nudi cay 2 pha sit dung tac nhan soc mudi, trong 24 gid dau, tao dugc lay 8 gid/lan de quan sat sy thay ddi hinh thai te bao dudi kinli hien vi quang hgc. Nhudm te bao bang Nile Red de quan sat kha nang tich liiy lipid va soi dudi kinh hien vi huynh quang.

Sinh trudng ciia tao dugc danh gia thong qua mat do te bao, mat do quang d budc song 680 nm (ODesonm), ham lugng sac to (chlorophyll a, astaxanthin) va ham lugng protein ndi bao [14, 21, 22]. Ham lugng chlorophyll a, astaxanthin va ham lugng protein ngi bao dugc xac djnh theo cdng bd cua Dgng Dilm Hdng va nnk. (2010) [ 10].

Nhudra te bao tao bang Nile Red theo quy trinh ciia Doan & Obbard (2011) [6]: Bd sung 5 pi dung djch Nile Red (9 - diethylamino - 5H benzo [a] phenoxazine-5-1) cd ndng do 0,1 mg/ml trong aceton vao 5 ml djch tao. Hon hgp dugc vortex nhe va u trong tdi 10 phiit d nhiet dp phdng. Sau dd, cac te bao tao da nhudm bang Nile Red se dugc quan sat dudi kinh hien vi huynh quang vdi budc sdng kich thich tir 450- 490 nm.

Tach chiet lipid tdng sd va xac djnh ham lugng lipid tong sd theo phuong phap cua Bligh

& Dyer (1959) (I] vdi mgt sd cai ti^n de phii hgp vdi dieu kien phdng thi nghiem cua Viet Nam: can 0,1 g sinh khdi tao khd cho vao coi chay sir, bd sung them 0,2 g Na2S04 va nghien bdn hgp nay thanh bot mjn. Sau dd bd sung them 10 ml hon hgp dung mdi chloroform : methanol (ty le 2:1 (v/v)) va ngam trong 1 gid d nhiet dd phdng. Lgc hon hgp qua giay lpc Whatman sd I va thu djch trong. Ba sinh khoi dugc tiep tuc chiet vdi chloroform tir 2 - 3 lan

lam bay hoi n-hexan de thu hoi lipid, Sd lieu thi nghiem dugc xii ly biing phan mem Excel va xit ly thdng ke ANOVA d rauc y nghTa P < 0,05.

Nhu v^y, ndng do 0,8% NaCl giiip te bag tao H. pluvialis chuyen giai doan tir cac te bao dang sinh dudng sang dang bao xac nhanh hon, Dieu nay rat quan trgng khi ap dung cho quy trinh nudi cay 2 pha ddi vdi loai vi tao luc nay.

Ham lugng protein ndi bao cd xu hudng giam dan theo thdi gian nuoi d tat ca cac cong thiic thi nghiem. Ket qua sau 35 ngay nuoi, ham lupng protein ngi bao cdn lai deu nhd hon 100 pg/te bao, Ci cdng thiic ddi chung, ham lugng protein ndi bao giam 1/2 lan so vdi gia Irj ban dau. Trong khi dd, bam lupng nay chi giam 1/4 lan so vdi gia trj ban dau d cae cdng thiie eo bo sung NaCl (hinh 5).

Cung vdi sy thay ddi ve ham lugng sac to va protein ngi bao thi ham lugng lipid tdng so cung cd su bien ddi trong qua trinh nuoi. Ket qua cho thay, ham lugng lipid tdng sd cd xu hudng tang dan trong qua trinh nudi. Khi cac te bao chuyen sang giai doan bao xac va tich Iuy astaxanthin thi dong thdi cd su tang ve ham lugng lipid tdng so, 0 thdi diem ban dau, ham lupng lipid chiem 13,54 ± 0,23%) trgng lugng khd-TLK va sau do tang dSn len 21,34 ± 0,35%. TLK d ngay thir 31.

Ket qua nay hoan toan tuong ty vdi eac ket qua ciia cac tac gia khac da cdng bd tren the gidi [4, 5, 16, 19].

Nhu vay, viec bd sung NaCl vdi cac nong do khac nhau da anh hudng len sinh truong va kha nang tich liiy astaxanthin d tao H. pluvialis.

Quy trinh nudi cay hai pha trong dd pha ragt nudi cay tag duoi dieu kien tdi uu de dat raat do te bao cue dai, sau dd chuyen sinh khdi tao vao pha thir 2 vdi cac dieu kien thuan lgi cho viec tich Iuy astaxanthin da dugc chimg ramh ia md hinh hieu qua di thu dugc sinh khdi H. pluvialis giau astaxanthin [18]. Trong thi nghiem nudi c§y 2 pha vdi pha II sdc mudi nay, chiing tdi da ti6n hanh theo diing quy trinh tren. Trude het, tao dugc nudi cay dudi dieu kien tdi uu cho den khi tao bat dSu di vao pha can b5ng vdi mat do tg bao eye dai la 28 ^ lO'' TB/ml va ham lugng

Theo cdng bo cua Boussiba & Vonshak (1991) [3], khi tao H. pluvialis gap dieu kien bat lgi nhu ndng do raudi cao thi te bao tao se cd ca ch6 ty bao ve bang each ngimg sinh trudng hoan loan, d bao chuyen tir trang thai sinh dudng, cd mau xanh, chuyen ddng sang dang bao xac, mau dd va tich Iuy lugng Idn astaxanthin trong te bao. Trong nghien cuu nay, chung tdi cung quan sat thay sy thay doi hinh thai cita te bao tao khi cam ung bang ndng dp

Ket qua chi ra d hinh 8 cho thay, trong 10 ngay ciia pha 1, mat dp te bao cd xu hudng tang nhe va dat gia tri cao nhat la 31 x IO"' TB/ml d ngay thii 5 sau nudi. Sau 10 ngay nudi, NaCl se dugc bd sung vdi ndng dd 0,8%i vao mdi trudng nudi d6 kich thich t6 bao tao tich liiy astaxanthin Luc nay, mat do te bao tao trong pha II cd sy sai khac ro ret giira cdng thirc ddi chimg va thi nghiem. O cdng thiic ddi chimg, mat do te bao cd xu hudng duy tri on djnh trong qua trinh nudi. Ngugc lai, d cdng thirc thi nghiem, khi cam ung bang ndng dp mudi cao se ire che sinh trudng cua tag, mat dd te bao cua cdng thite thi nghiera lai giam 2,5 lan so vdi gia

tri mat do te bao d§u pha II sau 22 ngay nudi.

Su thay ddi ve ham lugng sac td astaxanthin va hara lugng lipid tong sd trong nudi cay d pha II dugc trinh bay d hinh 9 va 10. Ket qua chi ra tren hinh 9 va 10 da cho thay, ham lugng astaxanthin/te bao ciia cdng thirc ddi chung va thi nghiera ed xu hudng tang dan trong ca pha 1 va pha II, Tuy nhien, khi cd su dyng tac nhSti sdc mudi, ham lugng astaxanthin/te bao tang dot bien. Sau 6 ngay sdc mudi, ham lugng sac td astaxanthin tang tir 5 pg/TB len 37 pg/TB.

Ham lugng lipid tdng sd eua tag d pha 1 cd xu hudng tang nhe tit 13,54 ± 0,23% TLK d 0 ngay dgn 14,92 ± 0,34% TLK d ngay thir 10. Sau khi chuyen tao sang pha II dudi dieu kien sdc mudi, ham lugng lipid tdng sd tang nhanh, tu 14,92 ± 0,34%, TLK len 17,14 ± 0,26%, TLK sau 6 ngay.

Microalga Haematococcus pluvialis Flotow is one of the natural sources of astaxanthin, a pigment widely used in aquaculture, food, pharmaceutical and nutraceutical industries. In the present study, we investigated the effect of high NaCl concentrations on growth and astaxanthin accumulation in the microalgae. Growth was observed under different salt concentrations (0,8%, 1 5% and 2,5% NaCl) according to two - phase culture mode. The obtained results have indicated that the high NaCl concentrations caused an increase in carotenoid content per cell and a decrease m the algal growth. The best carotenogenic condition by addition of salt was obtained at 2,5% NaCI under high temperature. Astaxanthin content per cell increased 4.8 folds in comparison to the initial value, from 10 pg.celf' to 48 pg.celf' after 15 days of salt stress. We observed that the accumulation of total lipid content was correlated with an increasing in astaxanthin content. The total lipid content of control was 15 ± 0,35% of dry cell weight whereas that of cell exposed to salt stress was 19% ± 1,3% and 26 ± 0.12% at 1.5% and 2.5% NaCl concentrations, respectively. This study will be an initial basis for the two phase culture process to obtain astaxanthin rich Haematococcus pluvialis biomass
