Summary
Human topoisomerase I (TOP-I) is the molecular target of a set of diverse anticancer compounds, including camptothecins, indolocarbazoles, indenoisoquinolines and
benzo[c]phenanthridines (BCP). Based on the crystal structure of DNA:TOP-I complex, 53 BCP derivatives were docked into the complex by DOCK 6.2 program. Docked BCP:DNA:TOP-l complexes indicated that BCPs tended to establish stronger interaction with DNA than with TOP-I. Moreover, a relationship between the biological activities and the docking scoring function was analyzed as well. Maybe, the information are useful for designing new BCP compounds of higher affinity.
