T 6 M T A T ; Benh dii thio dudng (DTD) li benh do rdi loan chuyin hda carbohydrate vdi dac trung la tSng glucose huyit man tinh. Trong dd enzyme glucose-6- phosphatase (G6Pase) cd ehu ydu a gan va thgn ddng vai trd quan trgng trong vi$c glQ- cin bSng glucose trong co the. Muc tieu chinh cua nghiin eCru nay li danh gii khi nang hogt dgng cua enzyme G6Pase cd trong gan vi thin cua chudt benh DTD duqc dieu tri bing eao chiit ethanol ti ciy li diea (Pandanus amaryllifolius). Chugt dugc gay benh DTD bing each tiem dung dieh alloxan monohydrate (AM) ndng do 130 mg/kg trgng lugng chufiL Chugt dugc xem la benh DTD khi co nfing do glucose huyet > 250 mg/dl, Sau khi chugt ti benh 7 ngay, ehugt benh DTD duge didu tri bang thudc thuong mai glucofast (150 mg/kg trgng lugng/ lan x 2 lln/ngay) hoac cao chill la cay la dua (400 mg/kg trgng lugng/ Idn x 2 lln/ngay) trong thdt gian 20 ngay.

2.3.3 Khao sat hogt dgng enzyme glucose-6-phosphatase a gan va than chugt

Sau 20 ngay thi nghiem, chugt duac giai phSu l l y gan va than de khao sat hoat dgng ciia enzyme G6Pase Gan (hoac than) sau khi giai phlu dugc l l y 500 mg nghiin trong 1 ml dung djch dem sucrose Ignh 0,25 M pH 6,5. Chat ddng nhat dugc ly tam d 4°C vdi tdc dp 3000 vong/phiit

Hogt dfing eOa enzyme G6Pase duge thyc hign theo quy trinh cOa Punitha ef al., [2] CO higu chinh nhu sau: Hdn hgp phan dng gdm 100 pl dung dich sucrose/EDTA 0,1 M pH 6,5 va 200 pl glucose 6 phosphate (G6P) 0,1 M duge pha trong 100 pl dung djch dgm citrate 0.1 M pH 6,5. Sau do hdn hgp phan dng dugc them vao 100 pl djch gan hogc thgn da dugc ddng nhat. Hdn hgp phan dng dugc l i e mgnh sau do dugc u d 37°C trong 30 phi^t. Phan dng dugc ket thuc bang 1 pl TCA/aseorbate (10%/2%. w/v) va tiip tyc duge ly tam vdi tde do 3000 vfing/phut trong 10 phut d nhidt do phdng. Sau khi ly tam, 100 pl djch trich dugc cho vao 400 pl dung dich A va 500 pl nudc khu khoang.

Dung dich A gdm 1,05 g axit ascorbic hoa tan trong 40 ml ciia hon hgp 0.2908 g postassium antymony tartrate, 12 g amoniummolypdate va 140 ml axit sulfunc dugc pha trong 1.3 lit nude khd khoang.

3. K £ T Q U A V A B A N LUAN 3.1 Hogt d6ng ciJa enzyme glucose-6- phosphatase trong gan cua chudt t h i nghigm udng cao la cay la dda

Glucose-6-phosphatase la enzyme eo chu yeu trong gan va thgn va la enzyme dong vai trd quan trgng trong vi$e chuyin hda carbohydrate. G6Pase xuc tac qua trinh chuyen hoa G6P thanh glucose va phosphate t y do. Nong dg phosphate vo CO duge tgo thanh do hogt dgng ciia enzyme G6Pase co trong gan chugt thi nghiem duge trinh bay d Bang 1. Ndng dfi phosphate duge tinh dya theo dudng chuan phosphate vd co y = 0,9953x + 0,0016 (R^= 0,9998).

Kdt qua trong nghien cdu nay cung tuang t y vdi nghifin edu cua Dai Thj Xuan Trang va cs., (2013) [4] khi nghien cdu higu qua uc chd enzyme G6Pase ciia cao chiet cay nhau {Morinda citrifolia). Ngoai ra Zhao et ai, (2011) [5] eung chung minh dugc ring enzyme G6Pase hoat dgng qua muc d chugt benh DTD. la nguyen nhan d i n den ndng dp glucose huyet tang cao. Cao c h i l l la cay la dda c6 kha nang dn ^ n h glucose huyit va giam hogt dfing cua enzyme G6Pase d chudt bfinh DTD trong nghien cuu nay la phii hgp vdi cac nghien cuu trudc day.

EFFECTS OF ETHANOLIC LEAF EXTRACT FROM PANDANUS AMARYLLIFOLIUS PLANT ON ENZYME GLUCOSE-6-PHOSPHATASE IN DIABETIC MICE

^College of Natural Sciences, Can Tho University Diabetes mellitus is characterized by abnormally high levels of glucose in the blood.

The chronic hyperglycemia is caused by different reasons. Out of these reasons, enzyme glucose-6-phosphatase (GOPase) which Is mainly presented in the livers and kidneys taking an imporiant role in controlling the blood glucose level. The objective is to estimate the activity of enzyme G6Pase in the livers and kidneys of diabetic mice treated with pandan (Pandanus amaryllifolius) ethanolie leaf's extract Mice were induced diabetes by intraperitoneal injection of alloxan monohydrate at concentration of 130 mg/kg body weight. After 7 days of diabetic incubation, mice having diabetes were orally administrated pandan leaf extract (400 mg/kg body weight x 2 times/day) or glucofast medicine (150 mg/kg body weight x 2 times/day). After 20 days of treatment, livers and kidneys of mice were used for determining the inhibitive ability on GOPase. The results showed that pandan leafs extract can significantly reduce the blood glucose concentration in diabetic mice after 20 days of treatment from 305.75 mg/dl to 125.5 mg/dl, a normal blood glucose level eventually. The inhibitive ability on GOPase of ethanolie extract from pandan leaves on livers and kidneys of diabetic mice was dramatically effective. The results also indicated that livers and kidneys of untreated diabetic mice produced 8.3257 mg/l and 3.0858 mg/l inorganic phosphate which significantly differed from those of control mice with 3.5902 mg/l and 1.4258 mg/l, respectively. By treating the diabetic mice with ethanolie extract from pandan leaves, the inorganic phosphate produced in their livers and kidneys significantly decreased with 2.4914 mg/l and 0.4119 mgA, repeetively. In conclusion, the extract of pandan leaves showed the anti diabetic property by inhibiting glucose-6-phosphatase in vivo. '
