This separates the clot from the side of the tube and separates out a good clear serum. If the holder of the free certificate finds any reactor in the annual test -- and - this _ often happens in the annual test twelve months after receiving the free certificate - he must pass the dean's tests before receiving another free certificate._ Detection of reactors, twelve months after. Tube testing began in the Union when poultry producers in the Ermelo district sent the first sets of samples to Onclerstepoort in 1937.

Looking at the progress of testing poultry for bacillary white diarrhea in the Union of r-3outh Ahiea, it was slow at first, but it has been maintained and there has been a gradual increase in the number of tests done annually was followed for the first five years by a point

At the beginning, peptone was also added, but it turned out that the growth of S. Before the growths from the Roux flasks are washed off, all condensation water is poured off. For washing the organisms from the medium, sterile physiological salt water solution, to which 0·2 per cent has been added. phenol, is used.

The organisms were washed off the medium with carbon-salt solution, the condensation water was retained. The condensation water was discarded and the organisms washed off with carbolic saline. The water of condensation was retained, the organisms were washed with carbolic saline, filtered through muslin, sedimented, and made into an unconcentrated suspension with fresh carbolic saline.

The water of eondensation was discarded, the organisms were washed with carbolic saline, not filtered, nm sedimented by centrifugation. The condensation water was retained, the cultures washed with carbonic saline, not filtered, sedimented and preserved.

These highly agglutinable single strains were used for sub<:heck test-retests performed with antigens composed of more than one strain of any S. In many cases, multiple reactions were elicited with single-strain antigens. These results were further verified by submitting the same post-active serum to multiple antigens of a single high-value strain.

Boxes, each containing 100 sera samples from a known infected farm, were taken and tested in parallel using two different single-strain antigens and a multi-strain antigen. Many birds missed by the antigens of several strains were left in flocks and within a month, i.e. Discussion.- (a) The best antigens are obtained by using a single injection to produce the antigen.

In most cases, more positive reactors are selected in poorly infected cl herds when antigens with one strain are used in preference than autigéens m ad e to more than one strain. In a small number of cases, an antigen with a lower value may cause more reactors than an antigen with a higher value.

Hutyra, Marek and iliannii~S'er (1838) mention bile and fioeculation and cite Gasman, Valley and Rettger who adjusted the pH of the antigen to 8·2 to 8·5 by adding an alkali and thus prevented these phenomena . The pH of most antigens was found to range from G·B to 7·6 using phenol red as an indicator with a Lovibond Comparator.

District ... considering that all my poultry and turkeys having been tested for bacillary white diarrhea and fowl typhoid by the Veterinary Division of the Department of Agriculture and Forestry acting under the direction of the Director of Veterinary Services, agree :- 2. Blood samples from all my poultry and turkeys over 4 months of age will be taken for the laboratory at a reasonable time, in such a way as may be approved by a person authorized by the Director of Veterinary Services. 3. If the result of the first analysis of the blood samples is negative, I undertake to test all the untested young birds and turkeys that I have now, after they reach the age of 4 months and anyway until the end of next year. March.

If all the fowls and turkeys I have now pass this first test without the reactors being detected, I understand B.W.D. and Avian Typhoid test certificate will be issued. I then undertake to forward to the Director like specimens annually, or at such times as he may think necessary, of all my fowls and keys over 4 months old, whether previously tested or not. If the test reactors are found in any of the fowls or turkeys, I undertake to send the Director any blood samples he may request and whenever he may make a written request for them, understanding that I will not I have to bleed any bird more often than once every four weeks and I will not be called, to get the test certificate for the whole undivided flock of poultry and turkeys to pass more than 2 clean tests in a row.

To allow a State Veterinary Officer access at all reasonable times to any or all of my chickens and turkeys for the purpose of collecting samples of blood for verification purposes. i). That all additions to my flock of birds and/or turkeys will be isolated, banded (see Clause l) and tested when brought onto the premises, and, before being added to my flock, such birds and/or turkeys again will be tested, the retest t::> takes place 30 to 40 days after the first test - provided that the testing of additions to the herd will not be necessary if they come from a herd which, after being tested as provided in paragraphs 2 and 3 hereof, is certified to be free of reactors to the B.W.D. That the fee to be paid by me to the Director of Veterinary Services shall be one penny in respect of each sper.imen blood tested by him.

This is not to be confused with any fee payable to a person authorized by the Director of Veterinary Services to take blood samples as set out in Articles 2 and 3 or to any registered veterinarian. To deliver to the Government Veterinary Officer for destruction or disposal in such manner as he may deem necessary, all hens and/or turkeys reacting to the aforesaid tests, and not to remove them from his premises without the written consent of the Government Veterinary Officer, it being understood that not I shall claim damages in respect of the birds thus surrendered by me and destroyed or removed by him. Postage is paid by the owner on samples sent from the farm to the laboratory. I hereby confirm that during the last two tests no reactors for the a-head test were found in the associated chickens and turkeys. As soon as the erum is loosened, the box of tubes can be removed and packed for shipment to the laboratory.
