Lampiran 2: Skema Kerja Pembuatan Preparat Histopatologi

Fiksasi Spesimen

Pemotongan Spesimen

Merendam spesimen dengan larutan BNF 10%, perbandingan antara spesimen dan larutan 1:10. Waktu fiksasi minimal 2 hari

 Spesimen dipotong dengan ketebalan 0,3-0,5 mm

 Disusun kedalam tissue cassette, kemudian dimasukkan kedalam keranjang khusus (basket)

Proses Dehidrasi

 Dimasukkan hasil pemotongan kedalam mesin processor otomatis

 Diproses selama 18 jam

Vakum

 Penghilangan udara dari jaringan dengan menggunakan mesin vakum (59-60°C)

 Proses selama 30 menit

Mencetak Parafin

 Jaringan spesimen dimasukkan kedalam setiap cetakan

 Parafin dituangkan kedalam cetakan, dibiarkan membeku diatas mesin pendingin

 Kemudian blok parafin dilepas dari cetakan

Memotong Jaringan

 Blok dipootong dengan menggunakan mikrotom dengan ketebalan 3-4 µm

 Potongan diletakkan di permukaan dalam floathing bath bersuhu 40°

 Irisan dirapikan, diletakkan di kaca object glass dan dimasukkan kedalam inkubator bersuhu 60°

Pewarnaan

 Pewarnaan dengan menggunakan pewarna Mayer’s Hematoxylin, kemudian diteteskan 1-2 tetes DPX, tutup dengan deck glass
