Xanthomonas axonopodis pv. glycines (Xag) is the c ausal age nt at bacte ria l pus tule dise ase of s oybeans. A non-pathoge nic mutant of Xag (M 715) was c ons tr ucte d e mploying tr ans pos on mutage nes is whic h s howe d similar epiphytic s urvival in planta to its wild type str ain (YR32). T he objective of this wor k was to ide ntify and to analyze ge nes involve d in pathoge nicity in Xag YR32. I nverse Polymerase Chain Reac tion (IPCR) was use d to is olate the DNA flanking transpos on inser tion. A 1.3 kb flanking DNA fragme nt was seque nced and analyze d e mploying BLAST pr ogr am to study homology, the position of transpos on inser tion and to pre dict the str ucture and f unc tion of the ge ne. One of the Ope n Reading Frames (ORFs) s hare d homology with inner membrane proteins (imps) of Xanthomonas axonopodis pv. citri
(GenBank accession No. NC 003919). Nor the r n blot analys is reve ale d that an imps ge ne was monocis tr onic and the size of imps mRNA in YR32 was slightly longer than in M 715. Reverse Transc riptase -PCR analysis demons trated that the imps transcr ipt in M 715 was muc h less abundant than I n the wild type YR32. Trans pos on (mini T n5Km'T p') was de ter mined to be ins erte d clos e to the e nd of C -terminal r egion of Imps ge ne and might be s uffic ie nt to destabili ze the imps transcr ipt I n M 715 and s o inf lue nce effectors trans por tation from Xag to plant ce ll.
