ABSTRACT
Bisphenol A (BPA), a xenobiotic estrogenic compound, has been widely used as a plastic monomer. The alterations of Sertoli and spermatogenic cells in prepubertal rats after short-term (6 and 9 hr) exposure to various concentrations of BPA in vitro were examined by TUNEL-staining and transmission electron microscopy. The testes of 20-day-old Sprague Dawley (SD) rats were cut into smaller pieces, and seeded in the medium. At 6 and 9 hr after administration of BPA at various concentrations (0, 10 nmol.mL-1, 1x10-2 nmol.mL-1, and 1x10-4 nmol.mL-1, respectively), the specimens were obtained for light and transmission electron microscopic observations. As a result, at 6 hr after exposure to BPA, the vacuolization within cytoplasm and the nuclear membrane rupture appeared in Sertoli cells. TUNEL-positive spermatogenic cells were also identified.

ALTERASI MORFOLOGIS PADA SEL SERTOLI DAN SPERMATOGENIK MENCIT SPRAGUE DAWLEY YANG DISEBABKAN OLEH
BISFENOL A: STUDI IN VITRO
ABSTRAK
Bisphenol A (BPA) yang merupakan suatu senyawa estrogenik senobiotik telah banyak digunakan sebagai monomer plastik. Dalam kaitannya dengan hal tersebut, alterasi sel Sertoli dan spermatogenik dari mencit yang diberi perlakuan pemaparan dengan berbagai konsentrasi BPA selama 6 dan 9 jam secara in vitro dilakukan dengan TUNEL-staining dan mikroskop electron transmisi. Testis dari mencit Sprague Dawley berumur 20 hari dipotong-potong menjadi berukukan kecil dan dibenamkan di dalam medium. Setelah 6 dan 9 jam pemberian BPA pada konsentrasi 0, 10 nmol.mL-1, 1 x 10-2 nmol.mL-1, dan 1 x 10-4 nmol.mL-1, specimen diamati dengan mikroskop electron transmisi dan cahaya. Hasil pengamatan menunjukkan bahwa setelah dipaparkan terhadap BPA terlihat adanya vakuolisasi dalam sitoplasma dan ‘rupture’ pada membrane inti sel. Sel spermatogenik TUNEL-positive juga dapat teridentifikasi. Alterasi sedemikian rupa cenderung meningkat secara gradual sesuai dengan perlakukan dosis dan waktu. Perlakuan BPA berhasil engidentifikasi sel-sel Sertoli dan spermatogenik apototik, dan sel-sel Sertoli dan spermatogenik nekrotik.
