ABSTRAK
Buah legundi (Vitex trifolia L.) diketahui memiliki flavonoid kastisin sebagai kandungan utamanya. Flavonoid kastisin dapat menyerap radiasi elektromagnet di daerah ultraviolet karena memiliki sistem aromatik sebagai kromofor dan gugus hidroksil sebagai auksokrom, sehingga dapat dianalisis dengan spektroskopi ultraviolet. Penelitian ini dilakukan untuk menetapkan flavonoid total dalam buah legundi dan menguji aktivitas antiinflamasinya. Analisis kandungan flavonoid total dilakukan dengan menggunakan spektrofotometer ultraviolet-tampak dengan bantuan pereaksi geser alumunium klorida dan kuersetin sebagai standar. Uji aktivitas antiinflamasi dilakukan dengan menggunakan metode induksi karagenan pada telapak kaki tikus dengan lima kelompok uji. Kelima kelompok uji ini terdiri dari kontrol negatif (PGA 2%), kontrol positif (asetosal 10 mg/kgBB), dan kelompok uji yang diberi ekstrak etanol buah legundi dengan dosis 500 mg/kgBB, 1000 mg/KgBB, dan 2000 mg/KgBB,. Hasil analisis flavonoid total buah legundi menunjukkan bahwa kadar flavonoid total yang dihitung sebagai kuersetin adalah 0,045% dari berat kering. Aktivitas antiinflamasi ekstrak etanol buah legundi ditunjukkan dengan inhibisi radang sebesar 21,44%; 30,78%; dan 41,46% berturut-turut untuk dosis ekstrak 500 mg/KgBB, 1000 mg/KgBB, dan 2000 mg/KgBB.

ABSTRACT
Legundi (Vitex trifolia L.) fruit has been known to contain casticin, a flavonoid, as its major constituent. This flavonoid absorbs electromagnetic radiation in ultraviolet region due to its aromatic system as the chromophore and its hydroxyl group as the auxochrome, therefore it can be determined by using ultraviolet spectroscopy. This research was conducted to determine the total flavonoid contained in legundi fruit and its antiinflammatory activity. The total flavonoid was analysed by using ultraviolet-visible spectrophotometer with aluminum chloride as the shifting-reagent and quercetin as the standard. Antiinflammatory activity assay was performed by using the carrageenan induced-method in five groups of rats. The tested groups were negative control (PGA 2%), positive control (acetocal 10 mg/kg of body weight), ethanol extracts of legundi fruit dose 500 mg/kg of body weight, 1000 mg/kg of body weight, and 2000 mg/kg of body weight, respectively. The total flavonoids contained in the extract which calculated as quercetin is 0.045% of dry weight.
