UJI EFEK HIPOGLIKEMIK EKSTRAK ETANOL DAUN DANDANG GENDIS (Clinacanthus nutans (Burm.f) Lindau) PADA MENCIT SWISS

Tujuan penelitian ini adalah untuk mengetahui efek farmakologi dari daun dandang gendis dalam menurunkan kadar glukosa darah mencit yang diinduksi aloksan. Bahan yang digunakan dalam penelitian ini adalah ekstrak etanol dari daun dandang gendis. 300 gram serbuk kering daun dandang gendis dimaserasi dengan etanol 96%. 18 ekor mencit jantan dibagi menjadi 6 kelompok yaitu kelompok I (kontrol negatif), II (kontrol positif), kelompok III (ekstrak dosis 58,8 mg/kgBB), IV (ekstrak dosis 117,6 mg/kgBB), V (ekstrak dosis 235,2 mg/kgBB) dan kelompok VI (kelompok pembanding glibenklamid 5 mg/kgBB manusia).

Aloksan diberikan pada hari pertama pengujian dengan dosis 70 mg/kgBB i.v.

Ekstrak etanol daun dandang gendis diberikan secara oral selama 7 hari berturut- turut dengan dosis 58,8 mg/kgBB (kelompok III), 117,6 mg/kgBB (kelompok IV) dan 235,2 mg/kgBB (kelompok V). Penurunan kadar glukosa diukur pada hari ke- 8 setelah perlakuan. Data dianalisis dengan ANOVA dan uji LSD untuk membandingkan kelompok II, III, IV, V dan VI. Hasil analisis data, ada perbedaan bermakna (P<0,05) antara kelompok II, VI, kelompok III, IV dan V.

Tetapi, tidak ada perbedaan bermakna antara kelompok VI, kelompok III, IV dan V. Hasil penelitian ini menunjukan bahwa ekstrak etanol daun dandang gendis dapat menurunkan kadar glukosa darah mencit yang diinduksi aloksan pada dosis 58,8 ; 117,6 dan 235,2 mg/kgBB.

The aim of this study was to know the pharmacological effect of dandang gendis leaves in decreasing blood glucose level on mice induced alloxan. Material use in this research was ethanolic extract of dandang gendis leaves. 300 g of powdered dried leaves of dandang gendis was macerated with ethanol 96%. The ethanolic extract obtained will be used for decreasing blood glucose level on mice induced alloxan. 18 male mice were divided into 6 group i.e group I (negative control), II (positive control), group III (extract dose 58,8 mg/kgBB), group IV (extract dose 117,6 mg/kgBB), group V (extract dose 235,2 mg/kgBB) and group VI (comparison group glibenclamid 5 mg/kgBB human). Alloxan was given on the first day of testing with dose 70 mg/kgBB i.v. The ethanolic extract of dandang gendis leaves be given orally during 7 consecutive days in the dose 58,8 mg/kgBB (group III), 117,6mg/kgBB (group IV) and 235,2 mg/kgBB (group V). Decrease of glucose levels were calculated from the 8th after treatment. The date were analyzed by ANOVA and LSD test to compare group II, VI, group III, IV and V.

Based on the data analysis, there were significance differences (P<0,05) between group II, group VI, group III, IV and V. But, there was no significance difference between comparison group, group III, IV and V. The result of this study showed thet ethanolic extract of dandang gendis leaves can decreased blood glucose level of mice induced alloxan at the dose 58,8 ; 117,6 dan 235,2 mg/kgBB.
