Regression analysis showed that the concentration of nitrogen in coral tissue was not significantly related to the density of nitrogen-fixing bacteria. The subject of the research was, among other things, the concentration of nitrogen in coral organisms and the density of nitrogen-fixing bacteria. The effect of nitrogen-fixing bacteria on the nitrogen concentration in corals was analyzed by regression.

The average density of nitrogen-fixing bacteria Nirobacter sp. is the las parameter observed in this research. However, there was no significant difference in the density of nitrogen-fixing bacteria Nitrobacter sp. The result of statistical tests showed that the effect of nitrogen-fixing bacteria showed that the concentration of nitrogen in the coral was not related to the density of Nitrosomonas sp.

The difference in the density of nitrogen-fixing bacteria is mainly caused by the environmental condition. However, nitrogen concentration in coral tissue is not affected by the abundance of nitrogen-fixing bacteria. However, in some cases, the availability of nitrogen in a coral reef ecosystem is limited. Nitrogen-fixing bacteria are mainly found in symbiotic reef-building corals as a source of nitrogen for coral growth (Wang et al., 2015). The least information is known about the processes and influence of nitrogen-fixing bacteria on the survival of coral communities.

This research aimed to study the nitrogen concentration in coral and analyze the influence of nitrogen-fixing bacteria on the nitrogen concentration in coral. The coral samples were stored in a cooler and identified in the laboratory for the density and concentration of nitrogen-fixing bacteria. Multivariate ANOVA was performed to analyze the difference in nitrogen concentration and abundance of nitrogen-fixing bacteria.

Regression analysis was performed using multiple regression to analyze the simultaneous effect of nitrogen-fixing bacteria and curve fitting to analyze the effect of nitrogen-fixing bacteria separately. Thus, it can be assumed that massive corals provided better support for nitrogen-fixing bacteria. Statistical analysis to assess the influence of nitrogen-fixing bacteria showed that nitrogen concentration in corals was not related to the density of Nitrosomonas sp.

Thus, it can be concluded that the concentration of nitrogen in the coral is influenced by some other factors, or involves some other factors as a moderator or as a cooperating factor. Nitrogen accumulation in massive and branching corals is suggested to be a growth rate effect between the two. Furthermore, nitrogen concentration decreases along with increasing depth (Alamaru, Loya, Brokovich, Yam, & Shemesh, 2009).

The symbiotic relationship between coral and the symbiodinium is very reliable in terms of nitrogen availability (Rädecker et al., 2015). The abundance of nitrogen-fixing bacteria indicates the nitrification rate provided by the community (Zulfarina, Rusmana, Mubarik, &. Santosa, 2017). The role of nitrogen-fixing bacteria is evident in the nitrogen-limited environment (Putnam et al., 2017).

Based on the result of the analysis, nitrogen concentration is higher in branching corals as an influence of its higher growth rate. Abundance of nitrogen-fixing, nitrifying, denitrifying and ammonifying bacteria in soil of tropical rain forest and oil palm plantations in Jambi.

The research was carried out in June – July 2019 in the Karimunjawa National Park area. The research stations were located in Karimunjawa Island, Menjangan Kecil Island and Cemara Kecil Island, while laboratory analysis was in Tropical Marine Biotechnology (TMB), Faculty of Fisheries and Marine Science Diponegoro University Semarang. The coral samples were stored in a cooler and identified in the laboratory for its density of nitrogen-fixing bacteria and the concentration of nitrogen.

The bacteria were isolated from the coral and placed in the enrichment media (nutrient agar). Statistical analysis showed that there was no significant effect of the source of coral samples and the types of coral on the density of Nitrosomonas sp. However, branching coral has higher Nitrobacter sp. density although the difference was not significant. The difference was obtained not only from the total average density, but also in the density of respective massive and branching coral types. Among the results of the assessment of nitrogen concentration as well as the density of Nitrosomonas sp.

When the sample was collected in the same size, the amount of coral polyps in the branching coral was more than the massive coral. However, the research found that there was no significant effect of the density of Nitrosomonas sp. The nitrification process took place in the surface of the polyps, but it mainly affects the availability of nitrogen in the environment.

The abundance of nitrifying bacteria is greater in shallow water (Smith, Damashek, Chavez, & Francis, 2016). In relation to the process of nitrogen fixation in Karumunjawa National Park, Nitrosomonas and Nitrobacter could play an important role in the remediation of the coastal environment. Although nitrogen-fixing bacteria exist in coral mucus or tissue, the bacteria are thought to be associated with Symbiodinium and not directly with corals (Lema et al., 2012).
