POTENSI FRAKSI n-HEKSANA EKSTRAK DAUN CENDANA (SANTALUM album, L) SEBAGAI ANTIINFLAMASI TERHADAP MODEL DERMATITIS KONTAK IRITAN PADA TELINGA TIKUS

YANG DIDINDUKSI MINYAK CROTON

ABSTRACT

Santalum album Linn (Cendana) has been traditionally used to treat inflammation. This research aimed to evaluate topical anti-inflammatory activity of n hexane extract of S.album leaves on croton-oil induced rat ear edema. The antiinflammatory activity test was done using 25 Wistar rats which divided randomly into five groups. Group 1 (negative control, croton oil 35%), group 2 (positive control, hydrocortisone drug 2.5%), groups 3 4, and 5 treated with 10, 20, and 40 mg/ear of n-hexane extracts respectively along with the croton oil. The thickness of rat ear-edema was measured manually using micrometre after 6 hours of observation. The topical application of n-hexane extract of S.album leaf at doses 10 and 20 mg/ear, and positive control hydrocortisone significantly reduced croton-oil induced rat ear oedema with percentage inhibition of 36, 52, and 73 % respectively. However, at a higher dose of 40 mg/ear, it did not significantly inhibit inflammation as compare to negative control. Histology analysis result also revealed that n hexane extract at doses 10 and 20 mg/ear, and hydrocortisone decreased significantly infiltration of leukocytes. Based on this result, it can be concluded that n-hexane extract of Santalun album Linn leaf exerted a great potency as topical antiinflammatory agent in a dose dependent manner.

INTRODUCTION

Iritant contact dermatitis [ICD], is an occupational skin diseases promoted by work materials or workflows such as solvents, detergents,degreasing agents, dust, and other substances [1]. Current antiinflammatory compounds available to treat ICD are both steroid and non streoid antiinflmmatory produced various side effects [2]. Thus, the search for a safer alternatives antiinflammatory drug derived from plants is necessary. n-hexane fraction of Santalum album L (cendana) extract demonstrated potent oral antiinflammatory activity in hind-paw rat oedema induced by carragenan therefore it is worth to evaluate whether it can also be used topically [3].

AIM

This research aimed to evaluate topical antiinflammatory activity of n-hexane fraction of cendana leaves extract on rats ear oedema induced by croron oil.

METHODS

Cendana leaves powder was extracted at room temperature using methanol to yield crude methanol extract. The crude methanol extract was fractionated into n-hexane. The n-hexane fraction was subjected to topical antiinflammatory test [4]. The topical anti-inflammatory activity test was done using 25 Wistar rats which were randomly divided into five groups. Group 1, negative control received only croton oil 35% in acetone, group 2, positive, control treated with standard drug (10 mg/ear

2 hydrocortisone cream 2.5%), groups 3,4, and 5 treated with different doses (10, 20, and 40 mg/ear) of n-hexane extract along with croton oil respectively. The thickness of rat ear-edema was measured manually using micrometre after 6 hours of observation.

RESULTS

The topical application of n-hexane extract of S.album leaf at doses 10 and 20 mg/ear, and positive control hydrocortisone significantly reduced croton-oil induced rat ear oedema with percentage inhibition of 36, 52, and 73 % respectively. The effeect of n-hexane extract at 20 mg/ear was comparable to drug hydrocortisone cream as statistical analysis demostrated insignificanly different.

However, at a higher dose of 40 mg/ear, it did not produced significant effect in inhibiting inflammation as compared to negative control. Histology analysis result also revealed that n hexane extract at doses 10 and 20 mg/ear, and hydrocortisone decreased significantly infiltration of leukocytes

CONCLUSION

It can be concluded that n-hexane extract of Santalun album Linn leaf exerted a great potency as topical antiinflammatory agent in a dose dependent manner.

SIMPULAN

 Fraksi n-heksan daun cendana yang diaplikasikan

secara topikal pada dosis 10 mg/telinga dan 20

mg/telinga mampu mengurangi ketebalan telinga tikus

yang diinduksi minyak croton dengan persen hambatan

inflamasi berturut-turut 36 % dan 52%, lebih rendah

dari kontrol positif obat hidrokortison (72%). Sedangkan

pemberian dosis 40 mg/telinga tidak menyebabkan

penurunan ketebalan telinga tikus secara signifikan

dengan persentase hambat hanya 18 %. Hasil analisis

histologi menunjukkan aplikasi fraksi n-heksana pada

dosis 10 dan 20 mg/telinga mampu menurunkan secara

signifikan infiltrasi sel radang.
