Lampiran 1 Pembuatan sel kompeten dengan perlakuan CaCl2(Sambrook et al. 1989)

Sebanyak 2 mL biakan E.coliDH5α umur 12 jam dikulturkan ke dalam 40 mL media LB. Kultur diinkubasi selama 2-3 jam pada suhu 37 °C sampai nilai Optical Density (OD600) mencapai 0.45-0.5. Selanjutnya kultur diambil sebanyak 1,5 mL dan disentrifugasi pada kecepatan 5000 rpm selama 15 menit. Pelet diresuspensi dengan 1 mL bufer transformasi. Sampel diinkubasi di dalam es selama 15 sampai 30 menit, kemudian disentrifugasi pada 5000 rpm selama 5 menit pada suhu 4 °C. Pelet diresuspensi kembali dengan 250 µL bufer transformasi dan disimpan dalam es.

RIWAYAT HIDUP

Penulis dilahirkan di Wonogiri pada tanggal 17 September 1989 sebagai anak ketiga dari pasangan Sutardi Hadi Wiyoto dan Kastini. Penulis menyelesaikan pendidikan menengah atas di SMA Negeri I Baturetno pada tahun 2007. Pendidikan sarjana ditempuh di Program Studi Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Negeri Sebelas Maret (UNS) Surakarta lulus pada tahun 2013. Pada tahun yang sama, penulis diterima sebagai mahasiswa Program Pascasarjana IPB di Departemen Bioteknologi dengan Beasiswa Pendidikan Pascasarjana Dalam Negeri (BPP-DN) yang diberikan oleh Direktorat pendidikan Tinggi (DIKTI) pada tahun 2013.

Selama perkuliahan penulis aktif dalam kegiatan organisasi kemahasiswaan Himpunan Mahasiswa Biologi (HIMABIO) UNS, Badan Eksekutif Mahasiswa FMIPA UNS dan MUTASI Study Club UNS. Penulis juga pernah menjadi asisten praktikum Ilmu Pengetahun Lingkungan, Kesehatan Lingkungan, Mikrobiologi, Genetika, Ekologi dan Fisiologi Hewan FMIPA UNS serta Farmakognosi jurusan Farmasi UNS. Penulis pernah aktif mengajar di salah satu bimbingan belajar di Surakarta pada tahun 2012-2013.
