ABSTRACT
Stevioside is a natural compound of terpenes. The sweetness of this compound about 250-300 times sweeter than sucrose (cane sugar), and low in calories. In Indonesia, this plant have not been developed and utilized in raw form only. The aims of this study were to determine the content of stevioside in crystals by high performance liquid chromatography (HPLC), solubility in water by measuring the refractive index, and the hypoglycemic activity of formulated stevioside sweeteners by glucose tolerance test methods in both Tawangmangu and Bandungan varieties. Blood glucose curve obtained was calculated area under curve using Graph 4.3.0.384. The data obtained were analyzed by a randomized block design (RBD) 6 groups and 4 replications. Testing differences between treatments used honestly significant difference (HSD) test with a significance level of 5%. The obtained results showed that stevioside crystal Tawangmangu varieties had stevioside content 52.54%, while the Bandungan varieties had higher content of 93.17%. Based on the refractive index, both crystals had almost the same solubility as sugar. Highest hypoglycemic activity of the sweetener was the sweetener derived stevioside Bandungan varieties dose 0.70 mg / kgbw in the amount of 67.55 ± 6.50%.
