UJI TOKSISITAS AKUT EKSTRAK ETANOL HERBA SELADA AIR (Nasturtium officinale R. Br. )
PADA ORGAN HATI MENCIT
ABSTRAK
Selada air (Nasturtium officinale R. Br.) termasuk suku Brassicaceae. Selada air memiliki khasiat dalam pengobatan sebagai antialergi, antidiabetes, antioksidan, antikanker dan pengobatan tuberkulosis. Tujuan penelitian ini untuk mengetahui toksisitas akut ekstrak etanol herba selada air.
Penelitian ini menggunakan metode eksperimental, dilakukan secara in
vivo menggunakan mencit jantan dan betina berjumlah 60 ekor, yang dibagi dalam
6 kelompok. Kelompok kontrol (K) diberi Na-CMC 0,5%, kelompok perlakuan (P) diberi ekstrak etanol herba selada air dengan dosis 50 mg/kg bb, 500 mg/kg bb, 1000 mg/kg bb, 2000 mg/kg bb dan 4000 mg/kg bb yang diberikan dalam dosis tunggal secara oral pada hari pertama. Dilakukan pengamatan terhadap gejala toksik, berat badan, berat organ relatif dan kematian mencit selama 14 hari, kemudian dilakukan pemeriksaan makropatologi dan histopatologi organ hati mencit. Data yang diperoleh dianalisis secara statistik dengan metode Two Way
Analysis of Variance (ANOVA) dilanjutkan dengan uji Post Hoc Tukey
menggunakan program SPSS (Statistical Product and Service Solution).
Hasil penelitian menunjukkan bahwa mencit jantan dan betina pada semua kelompok tidak menunjukkan gejala toksik pada uji panggung, uji katalepsi, uji urinasi, uji defekasi, uji salivasi. Hasil uji statistik menunjukkan bahwa ekstrak etanol herba selada air tidak berpengaruh terhadap perkembangan berat badan dan berat organ relatif mencit. Hasil pemeriksaan histopatologi organ hati mencit jantan dan betina menunjukkan bahwa pemberian ekstrak etanol herba selada air dosis 4000 mg/kg bb dapat menyebabkan ketoksikan pada organ hati, tetapi tidak menyebabkan kematian pada mencit jantan dan betina sehingga ekstrak etanol herba selada air termasuk dalam kriteria “Praktis Tidak Toksik”. Tidak ada perbedaan efek toksik pada mencit jantan dan betina. Dalam penelitian ini, LD50 tidak dapat ditentukan.

ACUTE TOXICITY TEST OF THE ETHANOLIC EXTRACT OF WATERCRESS HERB (Nasturtium officinale R. Br.)
IN THE LIVER OF MICE
ABSTRACT
Watercress (Nasturtium officinale R. Br.) including Brassicaceae family. Watercress has efficacy in the treatment of a hypo-allergenic, anti-diabetic, antioxidant,anticancer, and treatment of tuberculosis. The purpose of this research was to determine the potential for acute toxicity of ethanolic extract of watercress.
This research used experimental method, performed in vivo using male and female mice totaled 60 tails, which are divided into 6 groups. The control group (K) was given Na-CMC 0.5%, the treatment groups (P) were given ethanolic extract of watercress herb with dose 50 mg/kg bw, 500 mg/kg bw, 1000 mg/kg bw, 2000 mg/kg bw and 4000 mg/kg bw that given orally in a single dose on the first day. Observation of toxic symptoms, body weight, relative organ weight and death of the mice for 14 days, then examined the macropathology and histopathology of mice’s liver. Data were analyzed by using Two Way Analysis of
Variance (ANOVA) method and continued by Post Hoc Tukey test with using
SPSS (Statistical Product and Service Solution).
The results showed that male and female mice in all groups showed no toxic symptoms in stage test, catalepsy test, urination test, defecation test, and salivation test. Statistical analysis showed that ethanolic extract of watercress herb has no effect on the development of body weight and relative organ weight in mice. Histopatological examination of liver male ad female mice showed that administration the ethanolic extract of watercress herb with dose 4000 mg/kg bw can cause toxicity in the liver, but no cause of death in male and female mice, so that the ethanolic extract of watercress herb included in the criteria of “Practical Not Toxic”. There was no difference in toxic effect in male and female mice. In this research, LD50 can not be determined.
