Lampiran 1. Kurva Standar Protein

Pembuatan Reagen Bradford

Sebanyak 100 mg Coomassie Briliant Blue G-250 dilarutkan dalam 50 ml ethanol 95%.

Larutan tersebut kemudian ditambah dengan 100 ml asam fosfor 85%. Setelah

tercampur rata, larutan diencerkan dengan menggunakan ddH

O hingga 1000 ml.

Reagen harus disaring terlebih dahulu dengan menggunakan kertas saring sebelum

digunakan (Bollag & Stuart, 1991).

BSA (

Bovine Serum Albumin

) ditimbang sebanyak 100 mg dan dilarutkan dalam 100 ml

ddH

O hingga menjadi larutan stok BSA 1 mg/ml. Untuk membuat kurva standar

protein, dibutuhkan larutan protein dari konsentrasi 0 mg/ml; 0,1 mg/ml; 0,2 mg/ml; 0,3

mg/ml; 0,4 mg/ml; 0,5 mg/ml; 0,6 mg/ml; 0,7 mg/ml; 0,8 mg/ml; 0,9 mg/ml; dan 1

mg/ml. Dari masing-masing konsentrasi diambil sebanyak 0,1 ml dan direaksikan

dengan 5 ml reagen Bradford selama 5 menit. Larutan kemudian dispektrofotometri
