ABSTRAK
Kitosan merupakan salah satu biopolimer yang penting didunia dan juga hasil hidrolisisnya yaitu glukosamin karena terdapat banyak aplikasi di bidang medis, dan juga makanan. Analisis kitosan sering didahului dengan membuat derivatisasinya karena senyawa ini tidak memiliki kromofor. Penelitian ini bertujuan untuk mencari metode alternatif untuk analisis kitosan, baik secara langsung maupun yang sebelumnya dihidrolisis menjadi monomernya, yaitu glukosamin, dan direaksikan dengan fenilisotiosianat (PITC). Metode yang digunakan adalah metode spektrofotometri ultraviolet-sinar tampak orde nol dan orde dua. Hasil analisis kitosan secara langsung menunjukkan bahwa monomer kitosan, yaitu glukosamin, tidak bisa dianalisis tanpa diderivatisasi dengan PITC, karena molekulnya tidak memiliki gugus kromofor. Hasil analisis kitosan yang diderivatisasi dengan PITC menunjukkan bahwa monomernya, yaitu glukosamin, mempunyai serapan maksimum pada 272nm untuk spektrum orde nol, panjang gelombang zero crossing pada 272nm untuk spektrum orde satu, dan panjang gelombang serapan minimum pada 277nm untuk spektrum orde dua. Analisis kitosan terhidrolisis dihitung sebagai glukosamin memberikan hasil 77,92230% untuk spektrum orde nol dan 107,33464% untuk spektrum orde dua. Dari hasil penelitian ini, dapat disimpulkan bahwa kitosan dapat dianalisis dengan metode spektrofotometri ultraviolet orde nol dan dua.

ABSTRACT
Chitosan is one of the known biopolymer that is important to the world and the hydrolyzed product of chitosan, glucosamine is widely applied on the medical field and also food industry. The analysis of chitosan is always preempted with derivatization as this compound does not have a chromophores functional group. This research is conducted to find alternate method on analyzing chitosan either directly or hydrolyzed into its monomer, glucosamine, and is reacted with phenylisothiocyanate (PITC). Ultraviolet-Visible Spectrophotometry zero order and second order method was used. The direct analysis of chitosan resulted in the inability of analysis due to the lack of chromophore presence in the monomer, glucosamine, without forming a derivate with PITC. The result of the chitosan-PITC derivatization resulted that the monomer, glucosamine, shows maximum absorption at 272nm at zero order spectrum, the zero-crossing wavelength at 272nm for first order spectrum and the minimum absorption wavelength at 277nm for second order spectrum. The hydrolyzed chitosan analysis calculated as glucosamine yields 77.92230% for zero order spectrum and 107.33464% for second order spectrum. From the research, it is concluded that chitosan can be analysed with zero order and second order ultraviolet spectrophotometry method.
