ABSTRAK
Aspartam (L-alfa-aspartil-L-fenilalanin-1-metil ester) telah diketahui memiliki rasa manis. Turunan dari aspartil-dipeptida diketahui merupakan senyawa yang berpotensi sebagai pemanis (Laszlo Tarko et al., 2006). Pada penelitian ini dilakukan penggambaran fitur-fitur farmakofor dari turunan aspartil-dipeptida, agar dapat diketahui fitur-fitur farmakofor apa saja yang paling bertanggungjawab untuk memberikan rasa manis. Proses pemodelan farmakofor secara umum dapat dibagi menjadi empat tahap, yaitu meliputi pertama penyiapan data yang mencakup penggambaran 2D dengan ChemDraw Ultra, optimasi geometri dengan
Hyperchem 7.0, Common Feature Generation Pharmacophore dengan perangkat
lunak Discovery Studio, dan pembuatan training set. Kedua pharmacophore
generation dengan 3D QSAR Pharmacophore, ketiga analisis data, dan yang
keempat validasi model. Hasil penelitian ini dapat menunjukkan gambaran fitur-fitur farmakofor yang bertanggungjawab memberikan rasa manis yaitu gugus hidroksi dan karbonil berperan sebagai hydrogen bond acceptor (HBA), gugus amina sekunder berperan sebagai hydrogen bond donor (HBD), gugus benzen dan metil berperan sebagai hydrophobic (HY), dan gugus amina sekunder berperan sebagai positive ionizable (PI).

ABSTRACT
The aspartame (N-(L-α-Aspartyl)-L-phenylalanine,1-methyl ester), to possessing
a sweet taste has shown that the derivatives of aspartyl-dipeptides has the potential as sweeteners (Laszlo Tarko et al., 2006). So, it is suggested that a study of the sweetening level from the derivatives to be done. In this study, drawings of pharmacophore features of the aspartyl-dipeptides were done to discover which pharmacophore features are responsible for the sweetening ability. The pharmacophore modelling process consists of four levels. The first steps are the preparation of datas that cover the 2D drawing with ChemDraw Ultra, the geometrical optimization using Hyperchem 7.0, doing Common Feature Generation Pharmacophore using the Discovery Studio software, and the making of the training set. The second level is the pharmacophore generation with 3D QSAR Pharmacophore, the third is the data analysis, and the fourth is the model validation. Present study showed that the pharmacophore features responsible for the sweetening ability are the hydroxyls and the carbonyls as the hydrogen bond acceptor (HBA), the benzenes and the methyls as the hydrophobic (HY) group, and the secondary amines as the positive ionizable (PI) group.
