2. Phuong phdp

Nudi cay tiii ph&, khao sat su hmh thdnh va phat trien, kha nang tai sinh cua md seo theo quy trinh cua Lang, 2002 va cd cai tiin nhu sau:

- Mdt niia sd cdy trong cung ddng dugc tao r i trdn mdi trudng tao rd, mdt niia sd cdy cdn Igi sd dugc giu trong phdng. Cdy cd r i phdt triin dugc thich nghi trong dung dich Yoshida trong diiu kiin nhi^t dd phdng 3-5 ngdy trudc khi chuyin cdy ra trong trong nha ludi.

Mdi trudng tao md seo: N6 + 2 mg/l 2,4D + 60 g/lit sucroza; mdi trudng tdi sinh: MS +1 mg/l BAP + 1 mg/l NAA + 1 mg/l Kinetm + 30 g/1 sucroza ; mdi trudng nhan chdi: MS + 2 m g / l BAP + 30 g/1 sucroza; mdi trudng nhan nhanh: MS + 30 g/1 sucroza; mdi trudng tao rd: MS +1 mg/l NAA + 30 g/1 sucroza; mdi trudng thich nghi: dung djch Yoshida.

Anther culturability of rice is signilicantly different between crosses rice varieUcs. A doubled haploid (DH) population was estabbshed vi.i anther culture of some crossing from Fl on N6 medium, which had been shown particularly suitable for anther ciillurr of indica hybrids. Among these factors, the most important one is the genotypic difference. The results found that different rice species, or crosses quite differendy in response to anther culture. On the other hand, culture medium also strongly influences the andier culture response. The results that IR 64/OM 3536. OMCS 2000/ Jasmine M were which had high callus information rate. The results may be useful in the selection of parents with high response to anther culture for rice haploid breeding and in die establishment of permanent DH populations. There are 6 crosses Fl IR 64/Om 3536. IR 72/ OM 3536. OM 3536/ IR 73689-76. OM 3536/ IR 75997-159. OMCS 2000/ Jasmme .M and Tequng/ OM 3536 give generation good for yield and also quality.
