Day la nhung vat lieu dugc de xuat lam dong bd trong chuong trinh lai tao gidng chdng chiu dp dpc eua nhom.
Phat trien hai quan the hoi giao BCTv de thuc hien su sip xep thanh manh (fine mapping) tren ban do QTL da CO tu truac, ket qua cho thIy hai chi thi phan tu RM315 va RM210 dinh vi tren nhiem sic the so 1 va so 9 lien ket voi QTL chdng chiu dp doe cua nhom, thong qua gia tri chieu dai ve tuong doi (RRL). Nhung QTL gia dinh nay giai thich bien thien kieu hinh tir 11,85% den 13,8% trong quan the BCjFj ctia OMCS2000 / Kien Giang lpn; va tir 0,46% den 1,44% trong quan the BC.F, eiia OM1490 / AS996. (2001) da phat hien 5 QTL dilu khiln tinh chdng chiu nhom, dinh vi rli rIc tren 5 nhiem sic thi, vdi mot QTL chinh thtic dinh vi tren nhilm sic t h i sd 1. Nguyin vl etv.
(2002) tim fhly 10 QTL dinh vi tren 9 nhilm sic thi dilu khiln tinh chdng chiu nhom, tren co sd quin t h i luong boi k i p (DH) ciia td hgp lai CT9993 / IR62266. Nhirng phat hien nly khIng dinh ring: tinh chdng chiu do doc nhom II mot tinh trang phiic tap.
Nguyin Duy Bay vl etv. (2003) da sii dung 171 ddng Fg (ddng can giao tai td hgp = RILs) dugc chgn mot each nglu nhien tii 312 ddng ciia td hgp lai IR64 / Oryza rufipogon (ace. 106424 thu thap tai Trim Chim, dlt phln nang ciia Ddng ThIp Mudi) do tic gil va Ts. Bd me va con lai dugc thanh lgc nhom trong dung dich dinh duong Yoshida (Yoshida vl etv, 1976).
Nghiem thiic nhom dugc them vio la 0 va 40 ppm Al (tuong duong 1,48 mM), nhom boat dong Al'^* tu AICI3.6H2O d pH 4. Tdng ehilu dii bin dd II 1.755 cM, khoing cich trung binh gitra hai marker II 11,6 cM. Khi xem xet tinh trang chilu dai r l trong dieu kien cd xii ly nhom, tren nhilm sic t h i sd 3, quang giQa hai marker S1845-CD01395 cd mot tuong tac vdi khu vuc dugc xac dinh bdi RG653-RZ828 (tren nhilm t h i sd 6). Trong viec khai thIc chiln luge lira chgn chi thi phan tii (marker-assisted selection), QTL chinh lien kit voi CD01395 dinh vi tren nhiem t h i sd 3 cd the dugc xem nhu iing cii vien de ap dung viec chgn con lai cd gien chdng chiu do dgc nhom (BIy vl etv, 2003).

1. Xlc dinh vung QTLs gil dinh lien quan din tinh chdng chiu do ddc nhom (bd sung kit qua da dugc khIng dinh tren nhilm sic t h i sd 3 (BIy vl etv, 2003).
2. Xlc dinh chi thi phan hi d l khuyin cao trong chiln luge chgn gidng nhd microsatellites (ve tinh nhd).
II. VAT UEU VA PHUDNG PHAP l.Vatli$u
Thanh lgc hai quin t h i BC2F2 ctia cac td hgp lai OMCS2000 / Kien Giang Lgn, OM1490 / AS996.

2. Phuong p h i p
Thanh lgc do dgc cua nhom theo phuong phip thanh lgc ciia Buu va Lang (2003). Mot dong chgn 40 hat va cho vao trong tii sly tir 1-2 ngay (dl pha vo mien trang). Ngam cac goi hat trong nude Im trong 2 ngly va li cho nay m l m tren dia petri. Chon nhirng hat mgc m l m khoe, ddng deu d l cho k i t qui chinh xlc (dtmg nhirng hat moi nhii mlm d l cho hat vao tim mdp d l ding hem).
Thilt k l tim dp hinh khdi chu nhat: 40 cm x 30 cm X 2 cm (tuy loai khay nhua ma ta sii dung thilt k l cac tam dp cho phii hgp). Tren mdi tim dp, tao nhiing Id trdn dudng kinh khoing 2 cm. Mat dly ciia tim dp diing ludi cudc bao lai d l giCr hat liia. Chuyin cle hat gidng nay mlm vao cic Id (mdi Id cho vao mot hat) tren tam dp d l trong khay nhua. Sau khi cho vio dung dich dinh duong 30 ppm Al^* tii 2 - 4 tuln, tiln hinh do ehilu dii r l tdi da eiia 8 cay ma/mdi gidng. Phan tich phuong sai ctia 50 gidng ltia dia phuong, ddi voi RRL va gia tri trung binh dugc so sanh bing sir sai khac co y nghia nhd nhat (LSD) tren gidng chdng chiu AS996 lam ddi chiing.
Phuang phip dinh gi^ kilu gien nhd chi thi phan tii' SSR.
Chuong trinh MAPMARKER/EXP ver.3,0 (Lander va etv, 1987) dugc su dung de phan tich ban dd lien k i t gien voi h i m Kosambi (Kosambi 1944).
Chuong trinh MAPMARKER/QTL version 1.1 duoc su dung d l xlc dinh elc loci (dia diem) Inh hudng din nhung tinh trang sd lugng muc tieu tren cd sd phan tich bin dd each quang (Paterson va etv., 1988;
Lincoln vl etv., 1992), Sii dung LOD score = 3.0 lam nguong chgn lgc su hien dien QTL (cac dia diem tinh trang sd lugng) tren co sd khoang each tdng sd ciia bin do, va khoang each trung binh giii'a nhirng marker (chi thi phan tii) (Lander va Botstein, 1989).

III. KET QUA VA THAO LUAm

Su dung 50 gidng liia dia phuong chdng chiu phln dugc thu thap tai DBSCL d l danh gil, tim miu gidng phln iing chdng chiu vdi do dgc nhom. Cac gidng chdng chiu tdt bao gdm: Pokkah, Kien Giang Lgn. Bio cao truoc day ciia Vien Lua ddng bing song Clin Long cho rang, gidng liia ndi Ba Slo (Ddng Thap Muoi), Ning Phudc-(Tii Gilc Long Xuyen) diu thi hien tinh khIng do dgc nhom rat tdt. Gidng Ba Chiic va Tlu Niit phd bien a vung Tay Song Hau va Tii Giac Long Xuyen cung t h i hien tinh khIng nhom. Gidng Sa Quay thuoc nhdm liia mua som - phd hiln d An Bien (Kien Giang), trdng tren dlt nhilm man va than biin, cung dugc ghi nhan cd tinh khIng (Buu, 1987).
Kit qua thanh lgc (dilu kien lie e h l vdi 30 ppm Al) cho fhly cd 31 gidng bieu hien tinh chdng chiu nhom trong giai doan ma (62,0% miu thii nghiem).
Dd la: Kien Giang Lgn, Nip Mong Chim, Nang Me, Nang Thom Cho DIo, Nhen Thom (bing 1).
Mot sd gidng cd k h i nang mang gien chdng chiu do dgc ciia nhom nhung khong mang gien khang man nhu gidng: Mdng Chim, Tam Sic, Nang Thom Chg DIo, Nhen Thom, Ning Me, Hai Sung. Tuy nhien, gidng vira chiu man va vira chiu do dgc nhom dugc ghi nhan II: Kien Giang Lgn, Mahsuri, Tii Nguyen, Nhd thom, Ning Qudt, Trang Trdn. Day II nhirng ngudn vat lieu cho gien qui can chii y trong lai tao d l cai tiln trong ngan h i n g gien liia d ddng b i n g song Clin Long.

Cac chi thi phan tii SSRs lien k i t voi QTL g i l dinh dilu khiln tinh chdng chiu dd dgc nhom dugc tim fhly tren nhiem sic thi sd 1, 9 va 11 tren quin the RILg. Tir do giup cho viec xlc dinh markers trong viing muc tieu va xem xet nhting markers theo ngi dung sip xIp thanh minh - fine mapping tren quin t h i con lai dang phan ly.
Quin thi BC2F2 ciia OMCS2000/ Kien Giang Lgn duge dinh gil voi 190 ddng. Kit qui da hinh voi cac bang ddng hgp va di hgp tii (hinh 3 va 4) cho fhly RM315 ti-en nhilm sic thi sd 1 va RM201 ti-en nhilm sic the sd 9 hen kit k h i chat voi QTLs muc tieu dilu khiln tinh chdng chiu do dgc nhom. " Summary
Aluminum toxicity is considered as one of the main causes of low yield in rice. In this study, 50 traditional varieties were screened under Al toxicity stress of 30 ppm Al in Yoshida solution. Kien Giang Lon, Nep Mong Chim, Nang Me, Nang Thom Cho Dao, Nhen Thom, AS996 in the rice gene pool of CLRRI exhibited good donor for Al toxicity tolerance. Two BCjF, populations from the cross of OMCS 2000 / Kien Giang Lon and OM1490 / AS996 were used for fine mapping. The putative QTLs related to the relative root length (RRL) closely linked to RM315 and RM210 located in chromosome 1 and 9, respectively. Three QTLs namely qRRL-1, qRRL-9a, qRRL-9b, which were detected on chromosome 1 and chromosone 9 explained the phenotypic variation of 11,85% -13.8% in OMCS2000 / Kien Giang Lon.
