Tom tat: Ho Vinh Tan (thanh pho Vinh, Nghe An) la mpt frong nhiing noi tilp nhan ngudn nude thai cua thanh phd Vinh. Ket qua dieu tra cho thay nude hd da bi d nhilm hihi CO v^ 6 nhilm amoni. Doi vdi nganh tao luc (Chloropbyta), co 14 loai va dudi loai dugc tim thiy phin b6 trong h6 Vinh Tan. Tam chung tao luc thuoc cac loai Scenedesmus obliquus (3 chung), S. quadricauda (3 chung) \k Chlorella pyrenoidosa (2 chung) da dugc phan lap thanh cong, trong do, chung S. quadricauda VT-3C va S. quadricauda VT-5C c6 kha nang tich luy lipid tot nhat. Trong m6i tmdng AF-6, thdi gian nuoi ngan nhat de dat dugc mat do tl bao va ham lugng lipid cao nhit la 14 ngay; mat do tl bao cua chung VT-3C va VT-5C frrong ling la 138,39x10* vi 138,42x10* tl bao/mL; ty le lipid ciia chiing VT-3C va VT-5C dat tuong irng la 28,4% va 26,3% sinh khdi khd.
I. DAT VAN DE
Ngay nay, vi tao dugc xem la mdt frong nhiing ddi tuong tiem nang ifrig dung de san xuat diesel sinh hgc [5, 9] bdi vi cbiing cd kba nang tich liiy triacylglycerol vdi mdt ham lugng cao trong tt bao [12]. Mat khac, mdt sd loai nude thai co the su dung de nudi vi tao lay sinh khdi [10, 11]. Giai phap nay da dugc xay dung thanh chien luge dai ban cap qudc gia d mpt sd nude nhu Hoa Ky, Nhat Ban, Han Qudc, Ausfralia... ma diem khdi dau la phan lap, tuyen chpn dugc nhung chiing vi tao giau lipid, thich nghi tdt vdi dieu kien kbi bau dia phuang va vdi loai mdi tmong nude se su dung vao san xuat sinh khdi [9].
Tao luc (Chloropbyta) da dang ve thanh phan loai va pban bd rdng rai frong cac loai mdi tradng nude khac nbau [6]. Mdt sd loai giau lipid tbudc cac cbi Botryococcus, Chlamydomonas, Chlorella, Dunaliella, Scenedesmus... da dugc ngbien ciiu nudi frong cac loai nude thai khac nhau va cho nang suat sinh khdi, ham lugng lipid khac nhau [7, 11]. Trong nghien ciiu nay, cbiing tdi tap trang pban lap, danh gia va tuyen chgn mdt sd chung tao luc giau lipid tir hd cbiia nude thai dd thi Vinh Tan (thanh phd Vinh, Nghe An) de tao nguyen lieu cho cac nghien ciiu sau han theo hudng xii ly nude thai va san xuat diesel sinh hgc.
II. PHUONG PHAP NGHIEN CUU 2.L PhiroTig phap thu man

2.2. Phuong phap phSn tich chit lupng nirdc
Mdt sd chi tieu thiiy ly, tbuy hda dugc pban tich dua tbeo Quy chuan ky tbuat qudc gia ve chat lugng nude mat [4]. pH dugc do true tiep tai hien trudng bang may do pH. Oxy hda tan (DO) va nhu cau oxy hda sinh hgc (BOD5) dugc xac dinh theo phuong phap Winkler. Nhu can oxy hda bpc (COD) - Phuong phap kalibicromat. Amoni (NH4^ - Phuong phap so mau vdi tbudc thir Nesler. Photpbat (PO4"'") - Phuong phap so mau amonmolipdat. Chit rin la liing (TSS) - Phuang phap loc va can frgng lugng bang giay Igc dinh lugng.
2.3. Phuong phap quan sat va dinh loai loai tao luc
Cac tl bao tao luc dugc quan sat, md ta va do kich tbudc dudi kinh biSn vi quang hpc cd gin camera ket ndi may tinb va phin mem Motic Images Plus 2.0. Cac loai tao luc dugc dinb loai va sip xIp danh muc tbeo tai lieu pban loai cua Duong Diic Tien va Vo Hanh (1997) [6]. Mdi mau dugc lam 10 tieu ban; nhiing loai gap trong 8 den 10 tieu ban dugc quy uac la muc do gap nhieu (+++), 4 den 7 tieu ban dugc xem la miic dp gap tmng binh (++) va 1 din 3 tieu ban la mire do g^p it (+).
2.4. Phu-oug phap phan lap cac chiing tao luc
Mau tao thu tir bd Vinh Tan dugc nudi de tang sd lugng te bao bang each lay 150 mL mlu vao binh tam giac dung tich 500 mL, them 150 mL mdi trudng AF-6 [1], sue khi, dat dudi anb sang den neon vdi cudng dd chieu sang 3000 lux d nbiet dp phdng (25 - 30°C) trong 3 tuan. Sau do, te bao frong mdi gieng dugc chuyen sang nudi va bao quan trong mgt ong ngbiem cd mit nhya cbiia 10 mL mdi trudng AF-6; mdi dng ngbiem dugc xem la mdt ddng te bao hay mdt chung. Cac chung dugc quan sat va djnh loai.
2.5. Phuong phap nghiSn ciru sinh t r u ^ g cua cac chiing tao luc - Phuang phdp xdc dinh mat do ti bdo: Sd lugng te bao tai thdi diem xac dinh dugc dim tren budng dim hdng ciu va mat dp tl bao dugc tinb toan dua theo tai lieu cua Andersen R. A. (2005) [1].
- Phuang phdp xdc dinh sinh khoi kho: Sinh khdi te bao tai thdi diem xac dinh dugc thu hdi bing phuong phap li tam (6000 vdng/phut, 10 phiit), say khd d 60°C cho den khi frgng lugng khdng ddi (khoang 8 gid), can sinh khdi kbd bing can phan ticb.

Ong nghiem dugc li tam vdi tdc do 4000 vdng/phiit trong 5 phiit. 2.7. B 6 tri thi nghiem
- Thi nghiem doi vai cdc chung tdo Ivc duac phdn lap: Lay 30 mL gidng sau 10 ngay nudi ciia mdi chiing cbo vao binb tam giac dung ticb 500 mL cd chiia 300 mL mdi trudng AF-6 va dung mit day bing silicon cd 6ng dan kbi. Thi nghiem lap lai 3 lin dii vdi mdi chiing. Cac binh tam giac dugc sue khi lien tiic bang may tbdi kbi be ca; cbu ky chieu sang 12 gid sang : 12 gid tdi; cudng dp anh sang 3000 lux duoc cung cap bang den neon; dat d nbiet dp phong (25-30°C). Sau 16 ngay nudi, toan bd tl bao trong mdi binh dugc thu hdi de xac djnh sinh khdi khd, ddng tbdi tien banh tham dd kba nang sinb lipid cua chung.
- Thi nghiem doi vai mgt so chiing tdo liic dugc tuyin chon: Tbi nghiem dugc thiet ke tuong tu nhu tren vdi binh cau 5000 mL cd chiia 3000 mL mdi tradng AF-6 va dimg niit day cao su cd dng din khi, ty le gidng 10%. Mat dp te bao ciia mdi chiing dugc xac dinh sau khi nudi 0, 2, 4, 6, 8, 10, 12, 14 va 16 ngay. Kha nang sinh lipid dugc tham do bang hinb anh sau 10, 12, 14 va 16 ngay nudi.
- Thi nghiem nuoi sinh khoi di phdn tich hdm luong lipid tong so: De cd dugc lugng sinh khdi dii nhieu cho viec chiet lipid, cbiing tao dugc nudi frong 2 binh can dung tich 5000 mL tuong ty nhu tren. Sau 14 ngay nudi, dich nudi dugc ngimg sue khi, de lang trong 24 gid, hut bdt phan nude trong phia tren. Phan djch nudi dam dac dugc ii tam, say khd, tien banh chiet lipid va tinh toan ty le lipid tbu dugc so vdi sinh kbdi khd te bao.
III. KET QUA NGHIEN CUU VA THAO LUAN
3.1. Chat lircmg nirdc va thanh phan loai tao luc d ho Vinh Tan
Ket qua phan tich mdt sd chi tieu thiiy ly, thiiy hda ciia nude hd Vinh Tan vao thang 7 nam 2015 cho thay gia tri cac chi tieu vugt qua hoac khdng dat tieu cbuan ciia nude loai B| (QCVN 08:2008/BTNMT) diing cbo muc dich tudi tieu thiiy Igi hoac cac muc dich su dung khac cd yeu cau cbat lugng nude tugng tir (Bang I). Gia tri DO thap ban tieu chuan cho phep, BOD5 cao ban hai lan, COD cao hon ba lan; nhiing sd lieu nay chimg td nude hd Vinh Tan da bi d nhilm huu co. Ngoai ra, nude bd Vinh Tan con bi 6 nhilm amoni vdi ham lugng N H / cao bon bay lan tieu cbuan cho phep.

ISOLATION AND SELECTION OF LIPID-RICH STRAINS OF THE GREEN ALGAE IN VINH TAN MUNICIPAL WASTEWATER RESERVOIR
(VINH CITY, NGHE AN)
Vinh Tan reservoir (Vinh City, Nghe An Province) is one of places receving the municipal wastewater of Vinb City. Our investigation indicated that it was polluted with organic substances and ammonium. For the Chlorophytaphylum, 14 species and subspecies were found in Vinh Tan reservoir. Eight sfrains of the following species:
Scenedesmus obliquus (3 strains), S. quadricauda (3 sfrains) and Chlorella pyrenoidosa (2 sfrains) have successfully been isolated. Of these, tbe sfrains S. quadricauda VT-3C and S. quadricauda VT-5C showed the best abilities of lipid accumulation. In AF-6 medium, tbe shortest time of cultivation to obtain the highest cell densities and lipid contents was at the day 14 of tbe growth; accordingly, the cell densities of the sfrains VT-3C and VT-5C were 138.39xloSnd 138.42x10^cells/mL; tbe lipid contents of tiie sframs VT-3C and VT-5C were 28.4% va 26.3% of dried biomass, respectively.
