BAB I. PENDAHULUAN

A. Kesimpulan

1. Pada air dan lumpur kawah Sikidang Dieng terdapat 28 isolat bakteri yang memiliki aktivitas fitase, sehingga berpotensi sebagai sumber enzim fitase yang baru.

2. Identitas 3 isolat bakteri dengan aktivitas fitase tertinggi yang berasal dari kawah Sikidang Dieng berdasarkan gen 16S rRNA adalah Bacillus sp EN 6, Bacillus cereus EN 10, dan Bacillus cereus EN 16.

3. Karakterisasi bakteri penghasil enzim fitase dari air kawah Sikidang Dieng yang memiliki 3 aktivitas fitase terbesar yaitu:

a. Bacillus cereus EN 10 dengan aktivitas sebesar 0,32893 U/ml, memiliki suhu optimum 60ºC, memiliki pH optimum 4, aktivitas fitase dapat ditingkatkan dengan penambahan ion Ca2+ dan Mg2+, dan dihambat oleh ion Zn2+.

b. Bacillus cereus EN 16 dengan aktivitas sebesar 0,324953 U/ml, memiliki suhu optimum 50ºC, memiliki pH optimum 6, aktivitas fitase dapat ditingkatkan dengan penambahan ion Ca2+ dan Mg2+, dan dihambat oleh ion Zn2+ dan Fe2+.

c. Bacillus sp EN 6 dengan aktivitas sebesar 0,32182 U/ml, memiliki suhu optimum 60ºC, memiliki pH optimum 6, aktivitas fitase dapat ditingkatkan dengan penambahan ion Ca2+ dan Mg2+, dan dihambat oleh ion Zn2+ dan Fe2+.

B. Saran

Penelitian ini merupakan penelitian tahap awal, dan masih diperlukan tahap penelitian selanjutnya mengenai kloning gen dari isolat terpilih sehingga diperoleh sekuens DNA yang utuh, dan rekombinan DNA agar selanjutnya dapat dilakukan karakterisasi yang lebih terperinci dan dapat dimodifikasi sehingga menghasilkan enzim fitase yang memiliki stabilitas pada suhu tinggi dan dapat dimaksimalkan untuk aplikasi di bidang industri sebagai bahan suplemen pada pakan ternak.
