PROLIFERATOR-ACTIVATED RECEPTOR ALPHA (PPARα) DENGAN DAYA TAHAN OTOT (MUSCULAR ENDURANCE) PADA SISWA SEKOLAH SEPAK BOLA DI KOTA

Abstrak:
Pendahuluan: Gen Peroxisome proliferator-activated receptor alpha (PPARα) merupakan faktor transkripsi yang mengatur metabolism lipid, glukosa dan homeostasis energi didalam mitokondria. Variasi pada intron 7 pada gen PPARα berhubungan dengan fenotip kemampuan manusia. Hasil penelitian sebelumnya menyatakan homozigot GG lebih sering ditemukan pada atlit yang berorientasi pada daya tahan otot. Alel G berhubungan dengan peningkatan proporsi serabut otot tipe I dibandingkan pada serabut otot tipe II sedangkan alel C pada atlit yang berorientasi pada kecepatan atau daya ledak otot dan hipertrofi pada otot. Tujuan penelitian adalah untuk mengetahui hubungan polimorfisme gen PPARα dengan daya tahan otot pada siswa sekolah sepak bola di kota Medan.
Metode: Jumlah sampel 77 orang dan diambil sel bukal dan isolasi DNA. Polimorfisme gen PPARα intron 7 dilakukan menggunakan protokol PCR-RFLP dengan enzim TaqI. Tes Curl-Up digunakan untuk mengetahui daya tahan otot subjek
Hasil: Pada penelitian ini ditemukan genotip GG sejumlah 75 orang (97,4%) dan genotip GC sejumlah 2 orang (2,6%). Hasil tes Curl-Up mayoritas kategori sangat kurang sebanyak 51 orang (66,2%) dan kategori kurang sebanyak 12 orang (15,6%). Hasil uji Fisher menyatakan tidak ada hubungan yang signifikan (p=0,564). Pada populasi penelitian ini sesuai dengan asas Hardy-Weinberg Equilibrium (p=0,90).
Kesimpulan: Tidak terdapat hubungan antara polimorfisme gen PPARα dengan daya tahan otot (p=0,564). Daya tahan otot pada anaka- - anak sekolah sepak bola secara mayoritas sangat kurang. Rendahnya daya tahan otot mungkin disebabkan oleh faktor lainnya, seperti program latihan yang belum terstruktur, faktor gizi dan varian genetik lannya.

Abstract
Introduction: Gen Peroxisome proliferator-activated receptor alpha (PPARα) is a transcription factor that regulates lipid metabolism, glucose and energy homeostasis in the mitochondria. Variation in intron 7 on PPARα gene associated with the phenotype of human ability. Results of previous studies stated homozygous GG is more often found in athletes oriented muscular endurance. G allele is associated with an increased proportion of type I muscle fibers than in type II muscle fibers, whereas the C allele in the athlete-oriented speed or explosive power and muscle hypertrophy of the muscles. The research objective was to determine the relationship of PPARα gene polymorphism with muscle endurance in soccer school students in the city of Medan.
Method: Samples 77 people and taken buccal cells and DNA isolation. PPARα gene polymorphisms of intron 7 were performed using PCR-RFLP protocol with the enzyme TaqI. Curl-Up test is used to determine the subject muscular endurance
Result: This study found a number of GG genotype 75 (97.4%) and GC genotypes number 2 (2.6%). Curl-Up test results the majority of categories is very less as many as 51 people (66.2%) and less category as many as 12 people (15.6%). Fisher test results revealed no significant relationship (p = 0.564). In this study population in accordance with the principle of Hardy-Weinberg Equilibrium (p = 0.90).
Conclusion: There was no relationship between polymorphisms of genes PPARα with muscular endurance (p = 0.564). Muscle endurance in children - school children are the majority of football is very less. Low muscle endurance may be caused by other factors, such as exercise programs are not structured, nutritional factors and genetic variants.
