Teng) li loai nam g l ty nhien, hifn dang nhgn dugc sy quan tam die bift do cd nhieu hoat tinh sinh hgcquy, hua hen vugt frgi hon ca cac loai Linh chi: khang khdi u, uc chi tl bio ung thu, ting cudng khang thf, khang oxyhda [l-10].Ngay nay, ngudi ta daphan lap giong, thuin dudng v i nudi frdng thanh cdng.
Hifn nay, nam dugc frdng tgi Han Quic, Nhgt Ban, Trung Quoe va budc dau da dugc frdng d Viet Nam.Ciing mgt loai nhung dugc nudi trdng trong difu kien khic nhau,thanh phin va ham lugng cac boat chat frong nam cd thf thay ddi. 2. NGUYEN LIEU VA PHlTONG PHAP NGHIEN CtTU 2.1. Nguyen lieu
- Nim Thugng hoang duac mua td frang trgi sin xuat tai thanh phd Hd Chi Minhd dang qua thi da say khd va ddng gdi theo tifu chuin cua nha sin xuat. Ten Ioai da dugc nha cung cap giong va nhi san xuit xic nhan la Phellinus Ilnteus{Berk. et Curt.) Teng.
- Nim Dui ga (Pleurotus eryngii): gdm miu cd nguin gdc td Trung Quic vi miu dugc frdng tgi Da Lgt.
2.2. Chiet xuat cao ethaool, cao nirdc, cao PS, tinh chf PS va phSa doan
- Chilt xuit cao ethanol va cao nude; Mau cic loai nam dugcxay nhd,ngam chiet vdi ethanol (EtOH) 96° frong hai tuin dk tach logi cac hgp chit tan frong ethanol, cd duli dung moi, thu dugc cao ethanol. Sau dd, miu tiip tyc dugc chilt bing nude d 1OCC trong 4 gid. Qui trinh chilt dugc lap di lap Igi cho din khi dich chilt cudikhdng cdn polysaccharide (djnh tinh bing phenol-acid sulfiiric [I2]).Cd logidung mdi, thu dugc cao nude.
- Tich chilt va tinh chi cao polysaccharide (PS): Hda tan vdi nude cat viia dii, ket tiia vdi ethanol96° theo ^ If 1:5 (v/v), df qua dfm d-4''C rdi tien hanh ly tam thu lay PS tho. PS tho dupc tinh ehf bing phuong phip chilt vdi hdn hgp thudc thu Sevage (CHCIgiBuOH = 4:1, v/v) trong 30 phiit dk logi protein ty do.Qui trinh tinh chi logi protein niy dugc lip Igi nim lan den khi khdi lugng cao thu dugc gin nhu khdng dii. Sau khi Iy tam, phin tua lin lugt dugc nia sach vdi ethanol tuyft ddi vi acetone trude khi siy khd, thu dugc cao PSdatinh che.

2.3. Phutfng phap danh gia ho^t tinh khang oxy hoa
2.3.1. Xac dinh khi ning cho elefron theo lyc khang oxy hoi ting (total antioxydant capaci^) frong md hinh phosphomolybdenum
Lyc khing oxy hoa tdng ciia cac cao ethanol, cao nude, cao PS va cic phan dogn PS-E32, PS-
£48, PS-E68 tach chiet td nam Thugng hoing dugc danh gia theo phuang phap phosphomolybdenum ciia Prieto [2]. Phuong phap niy dya tren sy khu Mo (VI) vf Mo (V) bdi cac hop chit cd kha ning khing oxy hda frong mdi frudng acid, tgo thanh phuc phosphate/Mo

2.3.2. Danh gii lyc khang oxy hda qua kha ning bit glc ty do DPPH
Dya vao kha nang cic chit kliang hda chuyln hda glc ty do l,l-diphenyl-2- picrylhydrazyl (DPPH) thanh sin phim phin td lam cho dung dich chuyln tu mau tim sang vang. Pha dung dich DPPH ndng do 100 pM frong methanol ngay trude khi diing. Hdn hgp phin ling cd thi tich 3000 pL, gdm 1500 fiL miu khao sit d cac ning dp 100 fig/mL; 20 pg/mL; 4 pg/mL; 0,8 fig/mL va 1500 pL dung dich DPPH ndng dp 100 fiM. Cac hdn hgp phin ling dugc lie frong 1 phut va u d nhift do phdng frong 3D phut, rdi tifn hinh do quang d budc sdng 517 nm [9], Cdng thuc tinh:

SA DPPH (^): tic dyng bit gdc ty do (Scavenging Activity) DPPH cua miu.

Tit ca thi nghiem dugc lap Igi 3 lin. Tac dung bit gdc tu do dugc danh gia qua gia frj ICSO.MIU nim Diii ga (Pleurotus eryngii) glm miu cd ngudn gdc tii Trung Qudc vi man dugc frdng tai Da Lat ciing dupc khao sit df so sinh.
2.3.3. Xac dinh ciu tnie polysaccharide
- Methyl hda: Can 30 mg miu hda tan frong 2 mL dunethyl sulfoxide (DMSO) cho vao binh ciu hai cd cd lip hf thing hdi luu, khuiy tii frong 3 gid. Sau do them 30 mg NaOH tiep tuc khuiy tiJ frong 10 phut, lam lanh d 5''C, them tiip 17 mL (CH3)2S04 vio v i tiip tuc khuiy frong 16 gid. Chilt PS da metiiyl hda bing hon hgp CHCI3: HiO= 2:1 (v/v).
- Thuy phan: Liy 5 mg miu PS da methyl hda thuy phan vdi 3-4 mL dung dich acid CF3COOH 4M tgi 120°C trong 2 gid. Sau do, hdn hgp dugc lam mat vl nhift dg phong, rua bing CHjOH vi lim khd bing khi N2.
- Khu hda: Thfm 4 mL NaBHi 0,25M pha trong dung dich NHiOHIMvao mau sau khi thuy phan, giir d nhift dg phdng frong 30 phiit. Sau do thfm 5 mL dung djchacidCHsCOOH 10% trong CH3OH vio dl trung hoa lugng NaBHi du. Hdn hgp dugc thoi khd bing ddng khi N2.

- Acetyl hda: s i n p h i m da bi k h u h d a d u g c acetyl h d a v o l 1 m L hdn h g p (CH3CO)20;
C5H5N (pyridme) = 1:2 (v/v) tgi 100°C trong 2 gid. D f ngudi h d n hgp, them 5 m L CHsOHvalim khd bang ddng N2. Chiet san p h i m da acetyl h d a b a n g ethyl acetate [13].
M a u d u g c p h a n tich fren thiet bi s i c ky khi ghep khdi phd G C - M S Agilent 7890A sir d u n g cdt tach HP-5MS (30 m x 0,25 mm x 0 , 2 5 ^ m ) , khi mang H e v d i tdc do ddng 1 mL/phut.
C h u o n g frinh nhift d p : tii 120 "C nang nhift d o Ifn d i n 150 °C vdi tdc dp 10 "C/phut, tiip tgc nang nhift dp Ifn dfn 220 "C vdi tdc dp 3 "C/phut, sau d o nang nhiet d o d i n 280 "C vdi t i c do 10 "C/phiit. 4. KET LUAN
Trong nghifn cihi nay, kha ning khang oxy hda ciia cao polysaccharide tach chiet tu nim Thugng hoang (Phellinus linteus) hong tgi Vift Nam va caccao phin dognda dugc khao sat khi nang cho eleefron theo phuong phap phosphomolybdenum (Prieto, 1999) va khi ning bit glc ta do DPPH.Polysaccharide tich chilt tii nim Thugng hoing (Phellinus linteus) frdng tgi Vift Nam thf hifn kha nang cho electron cao ban nhilu so vdi cac loai nim thyc phim Dui ga (Pleurotus eryngii)tii Trung Quic va Vift Nam nhung thip hon so vdi curcumin. Dac bift kha nang cho hydro cua cao polysaccharide cua nim Thugng hoing (Phellinus linteus) cdn cao hon hin curcumm. Antioxidant activities of polysaccharide extracted from Thuong hoang mushroom (Phellinus linteus (Berk. Et Curt.) Teng) were studied The polysaccharide (PS) was extracted and purified to remove free protein. The PS was resolved in water and precipitated fy 32°, 48' vd 68 ethanol solutions, yield PS-E32. PS-E48, PS-E-68 fractions respectrvely.The antioxidant capacity of PS, PS-E32. PS-E4S, PS-E68. ethanol and water extract were evaluated by phosphomolybdenum method (Prieto. 1999) and DPPH free radical scavenging capacity. The results showed that the total antioxidant capacity of PS extracted from Thuong hoang mushroom is lower than total antioxidant capacity of curcumin. but the DPPH free radical scavenging capacity of PS is significantly higher than that of curcumin. Structure features of polysaccharides of ethanol 48'fraction (PS-E48) were investigated by monosaccharide compositions and glycoside linked poshion analysis.
The results indicated that PS-E48 were composed of D-mannose, D-glucose, D-xylose. D- galactose. D-arabinose with molar ratios of 1.00: 0.29: 0.12: 0.08: 0 06. respectively. The PS-E48 had D-mannan framework which contained main linkages I—*6-D-mannoside, branches contained linkages such as I^f6~D-glucoside and l—t6-D-galacloside.
