Hue tieu: Danh gia du'dc doc tfnh cap va doe tfnh ban tru'dng diln eiia che pham bgt acid amiii thiiy phan tir da ea tra, ea basa . Doi tu'dng va phu'dng phap nghien CLTU: bpt acid amln dddc danh gia doc tinh cap tren 42 chuot nhat trang va thiT doc tfnh ban tru'dng dien trSn 30 chuot cdng trang theo quy dinh chuan. Ket qua: nghien ciTU doe tinh cap ciia hot acid amin tren chuot nhat tring theo du'dng uong: LD50 = 14,17 ± 0,43 (g/kg TLCT/24h). Nghien eiTu doe tinh ban trddng dien, cho chu6t cong uong bot acid amin 42 ngay vdi lieu 140 mg/kg va 280 mg/ TLCT kg/24h.

SUMMARY

STUDY ON THE ACUTE AND SUBCHRONIC TOXICITY OF AMINO ACID POWDER FROM HYDROLYZED FROM SKIN OF PANGSIIDEA, PANGASIUS ON EXPERIMENTAL ANIMALS Objectives; Amino acid powder was hydnalyzed from skin of Pangsiidea, Pangasius. Materials and Methods: Amino acid powder was used to study on acute toxieitv in 42 mice and subchronic toxicity 30 rats by Standard Operating Procedure. Results: The result of acute toxicity of the amino acid powder: LDM

= 14,17 ± 0,43 (mg/kg/24h). The studying of the subchronic toxicity demonstrated that daily administering of the amino acid powder with doseage of 140 mg/kg and 280 mg/ TLCT kg/24h during 42 to rats, the base parameters have no signification for tiie weight of the body, Hematological indices (red blood cell, white blood cell, platelet and hemoglobin contents), hepatic indices (AST, ALT activity) and renal dicices (urea, creatinin levels) of rats after amino ado treatment were in normal biological values. Tissue images of the liver and the kidney are normal.

Gelatine la protein dddc tao ra td qua trinh thiiy phan collagen tiT da Idn, da bo, da ea, xu'dng. Cae acid amin ehii yeu cau tao nen gelatine la glycine, proline, 4-hydroxyproline va aspartie. Trong do acid amin glycine da dddc chiTng minh co tac dung ngan ngiTa sd lao hoa va tao ra do san chac cho da. Proline va 4- hydroxyproline la cae acid amin ly tu'dng cho viec phuc hoi khdp xu'dng, cd, cac mo va da. Axit aspartie rat quan trpng cho sd tang tru'dng mo va cd, tai tao te bao.

Hien nay vdi san lu'dng ca ea tra, ea basa nguyen lieu hang nam nhd hien nay, eac nha may che bien phai thai ra thi truSng hang tram nghin tan phe pham. Lu'dng chat thai nay eo the dude tan dung de die bien thanh thirc an chan nuoi, dau ea, dlesel sinh hoc, hoac xuat khau de lam nguyen lieu cho cong nghiep dude pham, my pham.

_^Viec tan dung phu pham da ea tra, ca ba sa de thiiy phan tao gelatine va cac ehiet xuat acid amin tao nguon nguyen lieu san xuat thu'c pham chiTc nang va my pham khong chi giai quyet du'dc 6 nhiem moi tru'dng ma eon tao ra eac san pham mdi eo nhieu tiem nang iTng dyng. Tuy nhien che pham acid amin tao dddc can du'dc danh gia tinh an toan tru'dc khi nghien ciTu va iTng dung d cac giai doan tiep theo. Chfnh vi vay trong nghien ciTu nay sau khi thuy phan da ca tra, ca basa bang gelatlnase va tao thanh che pham bpt acid amin chiing tdi tien hanh danh gia doc tfnh cap va doe tfnh ban tru'dng dien eiia che pham acid amin nay.

Bdl tddng nghien ciiii: Che pham bpt Acid amin thiiy phan tiT da ca tra, ca basa (co thanh phan - mg/lOOg mau: Glycine 3570 mg; Arginine 3024mg; Thrlonlne 2070mg; Alanine 70mg;

Methionine 3884mg; Lysine 2350mg) la san pham thuoc de tai ma so KC06.15/11-15.

Dpng vat thi nghiem: Chupt nhat trang dong Swiss tru'dng thanh, dii tieu chuan thf nghiem, khoe manh dat tieu chuan thf nghiem, trong

lu'dng 20g - 22g, Dong vat nghien eiTu do ban chan nuoi Hpe vien Quan y cung cap.

2.2. Phu'dng phap n g h i e n cu'u

^•2.1. Xac djnh ddc tinh cap theo du'dng udng: Xac djnh doc ti'nh cap theo du'dng uong dddc thdc hien theo phu'dng phap llvschitz P.G va cpng sd [1],[3].

2.2.2. NghiSn cud ddc tinh ban tru'dng dien cua ehS' pham bdt Acid amin theo du'dng udng tren chupt cdng trang [4],[5]

+ Lo chiTng (n=10): uong nu'dc cat lieu l,5ml/kgthetrpng/24h.

+ Lo diing thuoc lieu 1 (n=10): uong bot acid amin vdi the tfch l,5ml/kg dich thuoc tddng du'dng lieu 140mg/kg the trpng/24h

+ Lo diing thuoc lieu 2 (n=10): uong bot acid amin vdi the tich l,5ml/kg dich thuoc 'tu'dng du'dng 280mg/kg the trpng/24h

+ Chupt cong trang dddc cho uong nu'dc va thuoc thiT lien tuc trong 6 tuan lien tiep, moi ngay mot lan vao buoi sang

- Cic chi tiSu danh gii:

+ Theo doi tinh trang chung, hoat dong, an uong, trpng lu'dng cd the

+ Huyet hpc: hong cau, hemoglobin, bach cau, tieu cau.

+ Sinh hoa: AST, ALT, ure, creatinin.

Mo benh hoc: vao ngay thd 42, giet chupt, lay gan, than, quan sat hinh anh dai the gan, than va lam tieu ban darih gia hinh anh mo benh hpc.

- Thdi diem xet nghiem: Lay mau xet nghiem cae ehi so sinh hoa, liuyet hpc, ghi tai 3 thdi diem: xuat phat diem, sau 3 tuan, sau 6 tuan nghien cu'u. Thdi gian theo doi la 6 tuan,

Ket qua nghien cdu doe tfnh cifp eiia bpt acid amin tiiiiy phan tir da ca Tra, ca Basa tren dpng vat thdc nghiem cho thify vdi miTc lieu 11,00 g/kg TLCr/24h {mii'c lieu cao nhat chda xuat hien chuot chet) va vdi mire lieu 17,00 g/kg TLCr/24h {mu'c lieu thap lam chet 100% so chupt), da tinh du'dc LD50 ciia che pham bot Acid amin theo du'dng uong tren chupt nhat trang la: LDso = 14,17 ± 0,43 (g/kg TLCr/24h). Hhd vay, vdi mire lieu LD50 tim dddc tuWng du'dng quy do! ra lieu tren ngu'di la l,18g/kgTLa724h.
