Hiperurisemia merupakan suatu kondisi dimana kadar asam urat dalam darah melebihi kadar normal yang seharusnya. Daun tempuyung secara tradisional diketahui memiliki khasiat dapat menurunkan kadar asam urat dalam darah. Penelitian pendahuluan secara in vitro telah membuktikan bahwa ekstrak etanol daun tempuyung ini, mampu menghambat kerja dari enzim xanthine oxidase
sebesar 98,99% pada dosis 125 µg/ml. Melihat potensi tersebut, maka dilakukan penelitian guna melihat aktivitas antihiperurisemia secara in vivo terhadap mencit jantan galur swiss-webster dan mengetahui dosis optimal ekstrak etanol daun tempuyung yang dapat memberikan aktivitas antihiperurisemia. Pengukuran kadar asam urat darah dilakukan dengan menggunakan metode POCT (Point of Care Testing) selama 240 menit. Hasil penelitian memperlihatkan bahwa ekstrak uji dosis 500 mg/kg BB, dan 1000 mg/kg BB memberikan aktivitas antihiperurisemia dengan persen penurunan masing-masing sebesar 57,53% dan 55,81%, sedangkan dosis 250 mg/kg BB tidak memberikan aktivitas antihiperurisemia (P<0.05). Dosis 500 mg/kg BB merupakan dosis optimal dari ekstrak etanol daun tempuyung yang dapat memberikan aktivitas penurunan kadar asam urat darah mencit hiperurisemia.

iv ABSTRACT
Hyperuricemia is a condition where the uric acid blood levels in excess of normal levels should be. Tempuyung leaves are traditionally known to have the efficacy to reduce levels of uric acid in the blood. Preliminary in vitro studies have shown that the ethanol extract of tempuyung leaves, was able to inhibit the action of the enzyme xanthine oxidase by 98.99% at a dose of 125 µg/ ml. Seeing this potential, the study was conducted to evaluate the antihyperuricemia activity of ethanol extract of tempuyung leaves on male mice swiss-webster strain and to find the optimal dose of ethanol extract of tempuyung leaves that can give antihyperuricemia activity. Measurement of blood uric acid levels were measured by using the method of POCT (Point of Care Testing) for 240 minutes. Observations, showed that the extract dose of 500 mg/kg BW, and 1000 mg/kg BW gave antihyperuricemia activity with percent decrease respectively by 57.53% and 55.81%, while dose of 250 mg/kg BW did not provide antihyperuricemia activity (P <0.05). Dose of 500 mg/kg BW is the optimal dose of ethanol extract of tempuyung leaves that can provide activity to decreased blood levels of uric acid in hyperuricemia mice.
