EFEK IMUNOMODULATOR EKSTRAK ETIL ASETAT DAUN MAHKOTA DEWA (Phaleria macrocarpa (Scheff) Boerl.) TERHADAP RESPON HIPERSENSITIVITAS DAN TITER ANTIBODI SEL IMUN

ABSTRAK
Imunomodulator merupakan zat ataupun obat yang dapat mengembalikan ketidakseimbangan sistem kekebalan yang terganggu. Imunomodulator membantu tubuh untuk mengoptimalkan fungsi sistem imun. Tanaman mahkota dewa (Phaleria macrocarpa (Scheff) Boerl.) merupakan salah satu tanaman asli Indonesia secara empiris mampu menyembuhkan penyakit, seperti hepatitis, kanker, tumor, reumatik, alergi, asma, dan penyakit kulit. Tujuan dari penelitian ini adalah untuk mengetahui efek imunomodulator ekstrak etil asetat daun mahkota dewa pada mencit jantan.
Penelitian ini dilakukan secara eksperimental. Uji respon hipersensitivitas dengan mengukur pembengkakan telapak kaki mencit sedangkan uji titer antibodi ditentukan dengan hemaglutinasi. 25 ekor mencit dibagi menjadi 5 kelompok (CMC Na 0,5%, ekstrak etil asetat daun mahkota dewa dengan dosis 50 mg/kg BB, 100 mg/kg BB, 200 mg/kg BB dan levamisol 25 mg/kg BB sebagai kontrol positif) diberikan secara oral satu kali sehari selama 7 hari yang sebelumnya diinduksi dengan 0,1 ml sel darah merah sapi 1%.
Hasil penelitian menunjukkan bahwa ekstrak etil asetat daun mahkota dewa mampu meningkatkan respon hipersensitivitas dan titer antibodi secara signifikan dibandingkan kelompok kontrol (p<0,05). Dosis 100 dan 200 mg/kg BB menunjukkan efek yang lebih baik daripada suspensi levamisol 25 mg/kg BB. Hal ini menunjukkan bahwa ekstrak etil asetat daun mahkota dewa mempunyai efek imunomodulator terhadap respon hipersensitivitas dan titer antibodi sel imun mencit jantan.

ABSTRACT
Immunomodulators are substances or drugs that can restore an impaired immune system disorder. Immunomodulators help the body to optimize the function of the immune system. Mahkota Dewa (Phaleria macrocarpa (Scheff) Boerl.) is a native plant of Indonesia empirically able to cure diseases, such as hepatitis, cancer, tumors, rheumatism, allergies, asthma, and skin diseases. The purpose of this study was to investigate the immunomodulatory effect of Mahkota Dewa leaves ethyl acetate extract in male mice.
The study was conducted experimentally. Test hypersensitivity response was measured by swelling of the feet of mice while the test antibody titers was determined by hemagglutination. Twenty five mice were divided into 5 groups (CMC Na 0.5%, Mahkota Dewa leaves ethyl acetate extract with a dose of 50 mg/kg BW, 100 mg/kg BW, 200 mg/kg BW and levamisole 25 mg/kg BW as positive control) were administered orally once daily for 7 days previously induced by 0.1 ml of 1% cow red blood cells.
The results showed that, the Mahkota Dewa leaves ethyl acetate extract increased hypersensitivity response and antibody titers significantly as compared the control group (p <0.05). Doses of 100 and 200 mg/kg BW showed a better effect than the levamisole suspension 25 mg/kg BW indicating that the Mahkota Dewa leaves ethyl acetate extract revealed immunomodulatory effects on hypersensitivity response and antibody titer immune cells of male mice.
