UJI EFEK ANTIHIPERURISEMIA EKSTRAK ETANOL DAUN PEGAGAN
(Centella asiatica (L.) Urb.) PADA MENCIT JANTAN
ABSTRAK
Asam urat adalah produk akhir metabolisme purin. Kadar asam urat serum yang memiliki nilai di atas normal disebut dengan kondisi hiperurisemia. Hiperurisemia merupakan faktor resiko terjadinya arthritis gout, nefropati gout atau batu ginjal. Hasil penelitian sebelumnya menunjukkan pegagan memiliki efek anti inflamasi, antioksidan dan sitotoksik serta memiliki daya hambat terhadap enzim xantin oxidase secara in vitro, dimana enzim xantin oxidase merupakan enzim yang berperan dalam sintesis asam urat. Tujuan penelitian ini adalah untuk mengetahui efek antihiperurisemia ekstrak etanol daun pegagan pada mencit jantan hiperurisemia.
Pengujian efek antihiperurisemia dilakukan secara experimental dengan menggunakan alat pengukur kadar asam urat (Easy Touch® ) dan menggunakan
potassium oxonate 200 mg/kg BB sebagai penginduksi hiperurisemia yang
diberikan secara intraperitoneal dua jam sebelum pemberian perlakuan. Dua puluh lima mencit jantan dibagi menjadi lima kelompok, kelompok pertama sebagai kontrol negative yang diberikan CMC Na 1 % b/v secara oral, kelompok kedua sebagai kontrol positif yang diberikan allopurinol 10 mg/kg BB secara oral, kelompok ketiga sampai kelima diberikan ekstrak etanol daun pegagan dengan dosis masing-masing 200 mg/kg BB, 400 mg/kg BB dan 600 mg/kg BB diberikan secara oral, selanjutnya kadar asam urat diukur tiap satu jam selama empat jam pengamatan. Selanjutnya hasil yang diperoleh dianalisis menggunakan analisis variansi (ANAVA) yang dilanjutkan dengan uji Post Hoc Tukey HSD.
Hasil analisis menunjukkan bahwa semua dosis ekstrak etanol daun pegagan memberikan efek antihiperurisemia. Tetapi hanya dosis 400 mg/kg BB dan 600 mg/kg BB yang memberikan hasil yang tidak berbeda signifikan dengan pemberian allopurinol dosis 10 mg/kg BB.
Semua suspensi ekstrak etanol daun pegagan memiliki efek antihiperurisemia dengan dosis terbaik 400 mg/kg BB.

ANTIHYPERURICEMIC EFFECT OF ETHANOL EXTRACT OF GOTU KOLA LEAVES (Centella asiatica (L.) Urb.) ON MALE MICE
ABSTRACT
Uric acid is the final product of purine metabolism. When serum uric acid level higer than normal, it called hyperuricemia condition. Hyperuricemia condition is risk factor for arthritis gout, nephropathy gout or renal stone. Some research showed gotu kola has anti inflammatory effect, antioxidant effect and cytotoxic activity, gotu kola leaves also having inhibitory effect on the xanthin oxidase in vitro assay, wherein the enzyme xanthine oxidase is an enzyme that plays a role in the synthesis of uric acid. The purpose of this research was to study the activity ethanol extract of gotu kola leaves as antihyperuricemic agent on hyperuricemic male mice.
The examination antihyperuricemic effect was carried out by using uric acid tester (Easy Touch®) and potassium oxonate 200 mg/kg BW as inducer hyperuricemic given to the abdominal cavity one hour before treated. Twenty five male mice were divided into five groups, first group as a negative control treated carboxy methyl cellulose sodium 1% b/v orally; the second group as a positive control treated allopurinol 10 mg/kg BW orally ; the third to fifth groups were treated with ethanol extract of gotu kola leaves with each dose 200 mg/kg BW, 400 mg/kg BW and 600 mg/kg BW orally. Blood uric acid levels were measured every hour for four hours. Furthermore the result were analyzed by Analysis of Variance (ANAVA) followed by Post Hoc Tukey HSD test.
The result of examination showed that all doses ethanol extract of gotu kola leaves have antihyperuricemic effect, but only ethanol extract of gotu kola leaves doses 400 mg/kg BW and 600 mg/kg BW have the same effect with allopurinol 10 mg/kg BW.
All dose ethanol extract of gotu kola leaves have antihyperuricemic effect with the best dose is 400 mg/kg BW.
