EFEK EKSTRAK ETANOL DAUN BELIMBING WULUH (Averrhoa bilimbi L.) TERHADAP KONTRAKSI OTOT POLOS ILEUM
MARMUT JANTAN (Cavia porcellus) TERISOLASI ABSTRAK
Belimbing wuluh merupakan salah satu tanaman obat yang digunakan oleh masyarakat Blangpidie kabupaten Nangroe Aceh Darussalam sebagai antidiare. Penelitian ini bertujuan untuk mengetahui efek relaksasi dari ekstrak etanol daun belimbing wuluh pada otot polos ileum marmut terisolasi.
Tahapan penelitian ini meliputi persiapan bahan pengujian dan uji efek relaksasi menggunakan otot polos ileum marmut. Penelitian ini dilakukan secara
in vitro dengan menggunakan alat organ bath. Parameter yang diukur dalam penelitian ini adalah relaksasi otot polos ileum marmut terisolasi. Sebelum dilakukan pengujian, ileum marmut terisolasi diekuilibrasi selama 45 menit sampai diperoleh kondisi yang stabil didalam larutan tyrode dengan suhu 37oC yang diaerasi gas karbogen.
Hasil pengujian menunjukkan bahwa pemberian ekstrak etanol daun belimbing wuluh dengan seri konsentrasi pada otot polos yang sebelumnya diinduksi dengan asetilkolin menunjukkan penurunan kontraksi. Dengan adanya korelasi positif antara penurunan kontraksi oleh ekstrak etanol daun belimbing wuluh terhadap efek kontraksi ileum (r = 0,982). Ekstrak etanol daun belimbing wuluh pada konsentrasi 4mg/ml memiliki kemampuan yang tidak jauh berbeda dengan atropin sulfat 3x10-7M dalam menurunkan kontraksi otot polos ileum marmut terisolasi yang diinduksi oleh asetilkolin 2,91x10-5M (p > 0,05). Kontraksi yang ditimbulkan oleh asetilkolin dengan inkubasi ekstrak etanol daun belimbing wuluh menunjukkan hasil yang berbeda signifikan dengan kontraksi oleh asetilkolin tanpa inkubasi (p < 0,05), hal ini menunjukkan bahwa kemungkinan ekstrak etanol daun belimbing wuluh bekerja dengan menduduki reseptor muskarinik. Dapat ditarik kesimpulan bahwa ekstrak etanol daun belimbing wuluh memiliki efek merelaksasi otot polos ileum dengan kemampuan yang tidak jauh berbeda dengan atropin sulfat dan kemungkinan bekerja pada reseptor muskarinik.

THE EFFECT OF Averrhoa bilimbi L. LEAVES ETHANOL EXTRACT ON CONTRACTION OF ISOLATED GUINE PIG ILEAL (Cavia porcellus)
ABSTRACT
Averrhoa bilimbi L. is one of the medicinal plants as antidiarrheal that used by whole of people at Blangpidie Nangroe Aceh Darussalam. The aim of the study is determine the effect of relaxation the ethanol bilimbi leaves extract in isolated guinea pig ileal smooth muscle.
This study have some stages such as the preparing of material and testing of relaxation effect on guinea pig’s ileal smooth muscle. This research was conducted by in vitro methode using the organ bath. The parameter that measured in this research is the relaxation of isolated guinea pig’s ileal smooth muscle. Before the test, isolated guinea pig ileal were equilibrated for 45 minutes to obtain a stable condition in Tyrode solution at 37°C aerated carbogen gas.
The result is the ethanol extract of bilimbi leaves with serial concentrations of the smooth muscle induced by acetylcholine, that showed by decreases contraction. There have positive correlation between the decrease in contraction by the ethanol in bilimbi leaves extract with ileal contraction effect (r = 0.982). Extract ethanol in bilimbi leaves has concentration of 4mg/ml, that is not different with atropine sulfate 3x10-7M in reduced the contraction of guinea pig’s ileal smooth muscle induced by acetylcholine 2,91x10-5M (p > 0.05). Contraction of ileal induced acetylcholine with incubation of ethanol extract in bilimbi leaves showed that different significant results compare with the contraction of acetylcholine without incubation (p < 0.05), it showed that the possibility of the ethanol extract of bilimbi leaves has the mechanism of action muscarinic receptors. In conclusion, the ethanol extract of bilimbi leaves has the effect of relaxed the ileal smooth muscle. That are not different with atropine sulfate and the possibility mechanism of action muscarinic receptors.
