UJI AKTIVITAS ANTIBAKTERI DARI Lactobacillus sp. PADA BEBERAPA PRODUK MINUMAN YOGURT DI PASARAN
ABSTRAK
Yogurt merupakan produk yang diperoleh dari fermentasi susu dan susu rekonstitusi dengan menggunakan bakteri Lactobacillus bulgaricus dan
Streptococcus thermophilus atau bakteri asam laktat lain yang sesuai, dengan atau tanpa penambahan bahan pangan lain dan bahan tambahan pangan yang diizinkan. Lama penyimpanan dapat mempengaruhi viabilitas bakteri dan bakteri asam laktat yang digunakan sebagai starter mampu menghasilkan substrat antimikroba. Tujuan penelitian ini adalah untuk mengetahui pengaruh penyimpanan terhadap viabilitas bakteri pada sediaan yogurt yang diperiksa dan aktivitas antibakteri terhadap Staphylococcus aureus dan Escherichia coli.
Sampel yogurt diambil secara purposive sampling yang diperoleh dari Pasar Buah Pondok Indah di Jl. Setia Budi, Medan. Sampel terdiri dari sampel Yakult, Biokul dan Cimory. Pemeriksaan yogurt meliputi pemeriksaan viabilitas bakteri dalam yogurt selama penyimpanan 0, 5, 10, 15 dan 20 hari yang disimpan dalam refrigerator suhu 8oC dengan metode angka lempeng total dan aktivitas antibakteri terhadap Staphylococcus aureus dan Escherichia coli yang diperiksa pada hari ke-20 dengan metode difusi cakram.
Hasil penelitian menunjukkan adanya pengaruh viabilitas bakteri terhadap peyimpanan yaitu Yakult di hari ke-0 (271×108), hari ke-5 (241×108 cfu/ml), hari ke-10 (109×108 cfu/ml), hari ke-15 (83×108 cfu/ml), dan di hari ke-20 (50×108 cfu/ml), Biokul di hari ke-0 (133×108 cfu/ml), hari ke-5 (62×108 cfu/ml), di hari ke 10, 15, 20 tidak ada bakteri yang tumbuh, dan untuk Cimory dari awal sampai akhir penyimpanan tidak ada bakteri yang tumbuh. Untuk aktivitas antibakteri terhadap Staphylococcus aureus Yakult memiliki rata-rata zona hambat sebesar 7,1 mm, sedangkan Escherichia coli memiliki rata-rata zona hambat sebesar 9,5 mm. Biokul dan Cimory tidak mempunyai aktivitas antibakteri terhadap
Staphylococcusaureus dan Escherichia coli.

ANTIBACTERIAL ACTIVITY TEST OF Lactobacillus sp. IN SOME YOGURT DRINK PRODUCTS ON THE MARKET
ABSTRACT
Yogurt is a fermented product obtained from milk and milk reconstitution using the bacteria Lactobacillus bulgaricus and Streptococcus thermophilus or other lactic acid bacteria are suitable, with or without the addition of other foodstuffs and food additives are permitted. Storage time can affect the viability of the bacteria and lactic acid bacteria used as starter capable of producing substrates antimicrobial. The purpose of this study was to determine the effect of storage on the viability of the bacteria in the yogurt preparations were examined and antibacterial activity against Staphylococcus aureus and Escherichia coli.
Yogurt samples taken by sampling purposive obtained from Pasar Buah Pondok Indah street on Setia Budi, Medan. The sample consisted of Yakult, Biokul and Cimory. Examination yogurt includes examining the viability of the bacteria in yogurt during storage of 0, 5, 10, 15 and 20 days were kept in a refrigerator temperature 8oc by the method of total plate count and antibacterial activity against Staphylococcus aureus and Escherichia coli were examined on the 20th day with disc diffusion method.
The results showed the influence of the viability of bacteria to the storage that Yakult at day 0 (271 × 108), day 5 (241 × 108 cfu / ml), day 10 (109 × 108 cfu / ml), day 15 (83 × 108 cfu / ml), and on day 20 (50 × 108 cfu / ml), Biokul at day 0 (133 × 108 cfu / ml), day 5 (62 × 108 cfu / ml), at day 10, 15, 20 there is no bacterial growth, and to Cimory from the beginning to the end of storage no bacteria are growing. For antibacterial activity against Staphylococcus aureus
Yakult has an average of 7.1 mm zone of inhibition, whereas Escherichia coli has an average inhibition zone of 9.5 mm. Biokul and Cimory do not have antibacterial activity against Staphylococcus aureus and Escherichia coli.
