dalam Parafin I; campuran xylol-parafin (3:1), Parafin

II; campuran xylol-parafin (1:1), Parafin III; campuran

xylol-parafin (1:3), lalu Parafin murni

Embeding, dengan meletakkan organ hepar dalam kotak

berbentuk segi empat yang telah dipersiapkan

sebelumnya sebagai cetakan. Dituang parafin cair ke

dalam cetakan tersebut, lalu organ ditanam dan diatur

posisinya, kemudian diberi label. Dibiarkan sampai

dingin lalu disimpan dalam freezer. Kemudian

dilakukan penempelan blok parafin tersebut pada holder

ditempel pada holder menggunakan mikrotom sehingga

membentuk pita parafin dengan ketebalan 6 µm

Attaching,

dengan meletakkan pita parafin pada object

glass,

lalu dicelupkan ke dalam air dingin dan air

hangat, kemudian diletakkan di atas hotplate

selama

beberapa detik agar pita parafin lebih melekat pada

object glass.

Deparafinasi, dengan mencelupkan object glass ke

dalam xylol selama + 15 menit hingga parafinnya habis

Dealkoholisasi, dengan mencelupkan object glass ke

dalam alkohol bertingkat 100%, 90%, 80%, 70%, 60%,

50%, 40%, 30%

Stainning, dengan mencelupkan ke dalam pewarna

Hematoxylin Erlich selama 3-5 menit, lalu cuci dengan

air mengalir, kenudian dimasukkan dalam alkohol 30%,

50% dan 70%. Setelah itu dimasukkan dalam pewarna

Eosin 0,5% dalam alkohol 1-3 menit, lalu dimasukkan

dalam

aquadest

dan kemudian dimasukkan dalam

alkohol bertingkat 30%, 40%, 50%, 60%, 70%, 80%,

90%, dan 100%. Selanjutnya dimasukkan dalam xylol.

Mounting,

dengan menutup preparat menggunakan

cover glass dan Canada balsam.
