Mue dich cua nghien cihi nay la nhSm danh gia da dang di truyen cua 08 mau gidng liia HuySt rong, bao gom 04 mSu giong thu thap tai Quang Tri. Da dang di truydn cua cac miu giong liia da duuc danh gia a mue do phan tir voi cac chi tlii SSR. Ket qua phan lich da dang di truyen ciia 8 mhi giong lua Huyet rong vai 18 cap moi SSR da thu duoc 31 alen da hinh. So alen tren mot locut dao dong tir 2- 5 alen, voi gia tri Irung binh 2,6, K^t qua phan nhom dua trdn he sd tuong dong di truyen Dice da nhom cac miu giong thanh 2 nhom chinh, Ket qua danh gia cung cho thay 04 miu giong Huyet rdng thu thap tai Quang Tri da co sir tuong ddng v^ mat di truyen va co sii khac biet di truyen so voi cac miu giong Huyet rdng thu thap tai cac dia di^m khac. Cac mau gidng liia Huy^t rong chinh la nguon gien gia tri va la nguon vat lieu quy cho cong tac chon tao gidng liia dac san oViet Nam.

LSATVANDE
Huyet r6ng la giong lua quy, sinh trudng va phat trien tot tai cac viing dat ngap Iti, nhifem phen nhir viing Dong Thap Muai. Gao Huyet Rong mau do nau, be doi hat gao vin con mau hong ben trong, com ngon va co vi thom, la loai gao co gia tri dinh duong cao, kha nang chong chiu khi hau va khang benh tot. Hi6n nay, dien tich canh tac liia Huyet Rong con rat it.
Theo nghidn cuu cua cac nha khoa hpc, anthocyanin duoc giii lai trong lop vo lua mau do nau la mot chat co tiem nang chdng lai cac benh ung thu, tun mach va nhieu benh khac. Neu trong mot thang danh ra khoang muoi ngay an cac loai gao mau thay gao trang bmh thuong thi co the giiip ca th^ giai dpc, ha cholesterol, giam dau nhirc gan cdt, khop xuong ngon chan tay, thai dpc td trong gan, ngira ung thu.
Do Huy^t rong la gidng lua miia truydn thdng, canh tac tir nhieu thap ky, qua trinh canh tac lau dai da lam cho gidng liia nay giam chat luong va mat ddn thi trucmg. Do vay, vi^c phuc ti-ang giong lua Huyet Rong la rat cdn thiet.

Danh gia da dang di truyen nguon gien liia Huyet r6ng dia phuang dua tren kieu hinh va chi thi phan tii khong nhirng co y nghia trong viec bao ton cac mhi gidng liia dac san ma con CO dong gop quan trgng trong cdng tac chon tao gidng lua chat lupng cao.

- Vat Ueu thuc vat: 8 mlu gidng lua Huyet rdng (bang 1, hinh 1).
- Chi thi SSR 18 chi thi SSR cung cap boi hang IDT, My, phan bd rai rac tren 12 nhilm sac the cua he gien lua (bang 2).
- Cac hoa chat, dung cu, may moc dimg trong sinh hoc phan tir.

2.2.1. Phuong phap phan tich bing chi thi ph^
til SSR
ADN tdng sd dupe tach chiet theo phuang phap
"NaOH extraction" cua Wang (1993). Phan ung PCR duoc tien hanh tren may Veriti 96well Tliennal cycler vai tong the tich 15 pi, gom: 5 yl ADN, 0,15 \M mdi, 0,2 mM dNTPs, IX dich dem PCR, 2,5 mM MgClj va 0,25 don vi Taq TaKaRa. Dieu kien phan ung: 95^C - 7 phiit; 35 chu ky (94"C -15 giay, 55''C - 30 giay, 72°C - 1 phiit; 72''C - 5 phut) va giu mhi b 4''C. San pham PCR duac di&n di tren gel agarose 2,5% trong dem TBE, nhuom bang EtBr va dupe soi duai den UV.
2.2.2. Phuong phip phan tich sdh'du

3.1. K^t qua phdn tich da hinh ADN cdc mdu gidng lua Huy^t rong bang cac chi thi phan tu SSR
Trong nghien ciiu nay, da lua chpn 18 chi thi SSR phan bd rai rac trSn cac nhi&m sdc the cua he gen Ilia de danh gia sir khac biet giua cac mdu gidng Huyet rdng nhap npi so voi cdc mdu gidng Huyet rdng thu thap tai Quang Tri (Hinh 2).
K&t qua khao sdt da hinh ADN vdi 18 chi tiii SSR thu dupe ty 1^ da hmh kha cao, 12/18 chi thi SSR cho da hinh, chiem 65,67%. Tdng sd alen thu dupe tai 12 locut cho da hinh la 31 alen, sd alen dao dpng trong khoang 2-5 alen, trung binh 2,6 alen/locut. Trong do, chi thi RMl cho sd alen cao nhat voi 5 alen (Bang 3).
Ket qua phan tich cac chi sd chinh nhu tdn sd alen phd bien nhat va chi sd da dang PIC cua cdc locut SSR cho da hinh da cho thay tan so alen phd bien nhat dao dpng trong khoang tir 40 den 87,5%, tuang img vai chi thi RMl va RM22257, voi gia tri trung binh 61,02%. Chi sd da dang PIC cua cdc locut nghifen ciiu thay ddi tir 0,22 den 0,74, voi gid tn tnmg binh la 0,49. Gia tri PIC tinng binh phan dnh mice dd da dang chung cho tat ca cac locut nghien ciiu. Dieu nay chiing to nhiing chi thi SSR dupe sir dung trong nghien ciiu nay da cho kdt qua da dang kha cao giOa cac mdu gidng liia Huyet rdng nghien cuu, cd y nghia trong phdn tich di truyen cdy liia.

Summary
The purpose of this study was to evaluate the genetic diversity within 08 Huyet Rong rice accessions, including 04 accessions coUedted in Quang Tri province. Genetic diversity of the nee accessions was evaluated at the DNA level. Eight accessions were surveyed with 18 simple sequence repeat (SSR) markers reveabng the genetic relationship among the varieties. A total of 31 polymorphic bands were detected. The number of alleles per locus ranged from 2 to 5, with an average of 2.6. Cluster analysis based on Dice's sinulanty coefficients usuig UPGMA grouped the nee accessions into two major groups. The results showed that the 04 accessions coUedted in Quang Tn province showed genetic similanty among them and the genetic difference compared to accessions collected at other sites. The Huyet Rong rice accessions presents a valuable gene source and sufficient genetic background for specialty rice breeding in Vietnam.
