EFEK IMUNOMODULATOR FRAKSI N-HEKSAN DAUN MAHKOTA DEWA (Phaleria macrocarpa (Scheff) Boerl.) TERHADAP RESPON HIPERSENSITIVITAS DAN TITER ANTIBODI SEL IMUN

ABSTRAK
Imunomodulator membantu tubuh untuk mengoptimalkan fungsi sistem imun. Penggunaan tanaman obat semakin berkembang dimasyarakat, salah satunya adalah tanaman mahkota dewa (Phaleria macrocarpa (Scheff) Boerl) yang daunnya diduga memiliki aktivitas sebagai imunomodulator. Kandungan yang terdapat pada tanaman ini meliputi alkaloid, flavonoid, saponin, dan triterpenoid. Tujuan dari penelitian ini adalah untuk mengetahui efek imunomodulator fraksi n-Heksan daun mahkota dewa (FHDMD) pada mencit jantan.
Penelitian ini dilakukan secara eksperimental. Uji respon hipersensitivitas dengan mengukur pembengkakan telapak kaki mencit sedangkan uji titer antibodi ditentukan dengan hemaglutinasi. 25 ekor mencit dibagi menjadi 5 kelompok (CMC Na 0,5%, fraksi n-Heksan daun mahkota dewa dengan dosis 25 mg/kg BB, 50 mg/kg BB dan 100 mg/kg BB dan levamisol 25 mg/kg BB) diberikan secara oral satu kali sehari selama 7 hari yang sebelumnya diinduksi dengan 0,1 ml sel darah merah sapi 1%.
Hasil penelitian menunjukkan bahwa FHDMD dosis 25, 50, 100 mg/kg BB dapat meningkatkan respon hipersensitivitas dan titer antibodi sel imun mencit jantan dan terdapat perbedaan yang signifikan dengan CMC-Na 0,5% (p<0,05), respon hipersensitivitas dan titer antibodi yang paling baik ditunjukkan pada FHDMD dosis 100 mg/kg BB dengan perbedaan yang tidak signifikan dengan Levamisol 25 mg/kg BB. Hal ini menunjukkan bahwa FHDMD mempunyai efek imunomodulator.

IMMUNOMODULATOR EFFECT OF N-HEKSAN FRACTION OF MAHKOTA DEWA(Phaleria macrocarpa (Scheff) Boerl.) LEAVES ON HYPERSENSITIVITY RESPONSE AND IMMUNE CELLS ANTIBODY
TITER IN MALE MICE
ABSTRACT
Immunomodulator helps the body to optimize the function of the immune system. The utilization of medicinal plants progressively growing in the society, one of which is mahkota dewa (Phaleria macrocarpa (Scheff) Boerl). The leaves was reported to have an imunomodulatory activity. This plant contains alkaloids, flavonoids, saponins, and triterpenoids. The purpose of this study was to investigate the immunomodulatory effect of n-hexane fraction of Mahkota Dewa (HFMDL) Leaves in male mice.
The study was conducted by experimental. Hypersensitivity response test was measured by swelling of the feet of mice while theantibody titers test was determined byhemagglutination. Twenty five mice were divided into 5 groups (CMC Na 0,5%, n-hexane fraction of MahkotaDewa leaves at dose of 50 mg/kg BW, 100 mg/kg BWand levamisole at dose 25 mg/kg BW) were administered orally once daily for 7 days previously induced by 0.1 ml of 1% cow red blood cells.
The results showed that HFMDL dose 25, 50, 100 mg/kg BW increased the hypersensitivity response and antibody titer of the male mice and significantly different from those produced by CMC-Na 0,5% (p < 0.05), the highest activity on hypersensitivity response and antibody titer showed at EAEMDL dose 100 mg/kg BW which not significantly different with Levamisol®. This study proved that the HFMDL has immunomodulatory effect.
